摘 要:目的对达原饮进行拆方研究,探讨破结逐邪组、清热滋阴组、甘草组和全方组对大鼠免疫调节的作用机制,分析达原饮配伍合理性。方法制备脂多糖(LPS)诱导的大鼠模型,通过流式细胞仪(FCM)检测不同组方配伍的达原饮对脾淋巴细胞CD28^(+)、CD80^(+)分子表达的影响,酶联免疫吸附法(ELISA)检测培养上清液中白细胞介素-2,1β(IL-2,IL-1β)的含量,实时荧光定量聚合酶链式反应(PCR)和蛋白免疫印迹法(W B)检测脾淋巴细胞中PI3K mRNA和蛋白的表达。结果达原饮及各拆方组均能升高CD28^(+)水平、降低IL-2、PI3K mRNA水平;与对照组相比,除甘草组外,达原饮及各拆方组均能增强CD28^(+)、CD80^(+)的表达(P<0.05或P<0.01);与破结逐邪组、清热滋阴组及甘草组比较,全方组的IL-2、PI3K蛋白表达分别低于各拆方组(P<0.01),但在降低IL-1β、PI3K mRNA水平上,全方组只与清热滋阴组和甘草组有显著差异(P<0.01)。结论达原饮对免疫功能有显著的调节作用。各组分配伍对促进脾淋巴细胞CD28^(+),CD80^(+)分子的表达、降低细胞因子IL-2及IL-1β的分泌和下调PI3K mRNA和蛋白表达有差异化协同作用,总体上达原饮全方更具优势。OBJECTIVE To study the disassembled prescription of DaYuanYin(DYY),which includes breaking knot and expelling evil group,clearing heat and nourishing yin group,licorice group and whole prescription group,on the immunoregulation of rats,and analyze the compatibility rationality.METHODS The model of rats induced by Lipopolysaccharide(LPS)was used.The effects of different prescriptions of DYY on the expression of CD28^(+)and CD80^(+)molecules in spleen lymphocytes were tected by flow cytometry.Interleukin-2,-1β(IL-2,IL-1β)of the culture supernatantwere examined with ELISA.The expression of PI3K mRNA and protein in spleen lymphocytes were detected by real-time fluorescence quantitative PCR and Western blot.RESULTS DYY and the disassembled prescription group could increase the level of CD28+and decrease the level of IL-2 and PI3K mRNA;Compared with the control group,except for licorice group,DYY and each prescription group could enhance the expression of CD28+and CD80^(+),and the protein expression of IL-2 and PI3K in DYY group was lower than that in the other groups(P<0.05 or P<0.01);And compared with the three groups,the expression of IL-2 and PI3K protein in the DYY group was lower than that in the other groups(P<0.01),but there was significant difference in reducing the level of IL-1βand PI3K mRNA only between the whole prescription group and the heat-clearing and yin-nourishing group or licorice group(P<0.01).CONCLUSION DYY has a significant regulatory effect on the immunoregulation.The compatibility of each group has a differential and synergistic effect on promoting the expression of CD28+,CD80^(+)molecules in spleen lymphocytes,reducing the secretion of cytokine IL-2 and IL-1βand down-regulating the expression of PI3K mRNA and protein.Taking together,the completed DYY has more advantages.
