摘  要:试验旨在研究试剂A应用于牛基础成熟液时的体外胚胎发育效果以及牛卵巢常温保存的可行性。试验分为两部分,试验1为对比试剂B与试剂A作为牛基础成熟液对体外胚胎发育的影响,试验2为对比常温(23℃)与37℃保存卵巢对体外胚胎发育的影响,分别测定两对比条件下获得的卵母细胞数、卵裂率、囊胚率。结果显示:试剂A的囊胚率达到了19.04%,显著低于试剂B(P<0.05)。卵巢常温(23℃)保存时,其囊胚率可达21.75%,显著低于37℃保存条件下(P<0.05)。研究表明,试剂A可用于牛体外胚胎生产体系的建立,但效果与试剂B存在一定差距;卵巢可在常温条件下保存,但其效果亦不如37℃保存条件。The purpose of the experiment was to study the in vitro embryo development effect of reagent A when applied to bovine basal maturation fluid and the feasibility of storing bovine ovaries at room temperature.The experiment was divided into two parts,test 1 was to compare the effect of reagent B and reagent A as bovine basic maturation fluid on embryonic development in vitro,test 2 was to compare the effect of ovary preservation at normal temperature(23℃)and37℃on embryonic development in vitro.The number of oocytes,cleavage rate and blastocyst rate obtained under the two contrasting conditions were measured respectively.The results showed that the blastocyst rate of reagent A reached19.04%,which was significantly lower than that of reagent B(P<0.05).When the ovary was stored at room temperature(23℃),its blastocyst rate could reach 21.75%,which was significantly lower than that at 37℃(P<0.05).The experiment indicates that reagent A can be used for the establishment of bovine embryo production system in vitro,but the effect is different from that of reagent B.Ovaries can be stored at room temperature,but the effect is not as good as that at 37℃.
