摘  要:【目的】为了分析脂代谢相关的差异表达基因和代谢通路在伞裙追寄蝇Exorista civilis滞育调控过程中的作用。【方法】本研究利用高通量测序对伞裙追寄蝇滞育和非滞育蛹进行转录组学分析,通过生物信息学方法筛选出与脂代谢相关的差异表达基因,并进行了GO和KEGG富集分析。【结果】在GO富集分析中,差异表达基因共富集到48个GO条目中,而生物过程富集的差异表达基因数量最多。在KEGG富集分析中,成功注释的112个差异表达基因,其中48个上调表达,64个下调表达,涉及15条通路,主要包括甘油磷脂代谢、脂肪酸降解及甘油磷脂代谢等通路,富集在这些通路中的差异表达基因呈现不同的表达趋势。【结论】在脂代谢中脂酰辅酶A还原酶、硬脂酰辅酶A脱氢酶和磷酸甘油脱氢酶的差异表达分析可能对伞裙追寄蝇滞育期间的表皮形成、抗寒性提高及能量传递有着重要的影响。[Objectives]To analyze the roles of differentially expressed genes and metabolic pathways related to lipid metabolism in the diapause of Exorista civilis.[Methods]High-throughput sequencing was used to complete the transcriptomic analysis of diapausing and non-diapausing pupae.Differentially expressed genes related to lipid metabolism were screened with bioinformatic methods and analyzed using GO and KEGG enrichment.[Results]GO enrichment analysis indicates that differentially expressed genes were enriched to 48 GO terms and that the largest number were enriched by biological processes.KEGG enrichment analysis successfully annotated 112 differentially expressed genes involved in 15pathways,of which 48 were up-regulated and 64 down-regulated.Differentially expressed genes were mainly involved in the glycerophospholipid metabolism,fatty acid degradation and the glycerolipid metabolism.Differentially expressed genes enriched in these pathways showed different expression trends.[Conclusion]Fatty acyl-CoA reductase,stearoyl-CoA desaturase(delta-9 desaturase),and glycerol-3-phosphate dehydrogenase are differentially expressed in the lipid metabolism of E.civilis,and may therefore play an important role in formation of the epidermis,cold resistance and energy transfer,during diapause.
