摘  要:目的探讨大鼠心肌细胞使用不同浓度紫草素处理后的毒性作用。方法选取出生1~3d的新生乳鼠,取其心室的心肌细胞进行培养,采用MTT法检测不同浓度梯度(1×10^(-7)mol/L、1×10^(-6)mol/L、1×10^(-5)mol/L)紫草素处理心肌细胞48h、72h后的细胞毒性。结果MTT检测结果发现,细胞毒性实验作用48h、72h,紫草素浓度为1×10^(-5)mol/L时,心肌细胞均有显著毒性(P<0.05),紫草素浓度为1×10^(-7)mol/L、1×10^(-6)mol/L时,心肌细胞无显著毒性(P>0.05)。结论紫草素具有一定的细胞毒性,紫草素浓度小于1×10^(-6)mol/L时,未表现出明显毒性,当紫草素浓度为1×10^(-5)mol/L时处理心肌细胞48h、72h后,对大鼠心肌细胞有明显毒性。Objective To explore the toxic effects of rat cardiomyocytes treated with different concentrations of shikonin.Methods Select neonatal rats born from 1 to 3 births,and culture the cardiomyocytes of their ventricles.and the cardiomyocytes were treated with shikonin in different concentration gradients(1×10^(-7) mol/L,1×10^(-6) mol/L,1×10^(-5) mol/L)by MTT method for 48h,Cytotoxicity after 72h.Results Using MTT test results,it was found that the cytotoxicity test effect was 48h,72h,and when the shikonin concentration was 10^(-5) mol/L,the cardiomyocytes had significant toxicity(P<0.05),and the shikonin concentration was 1×10^(-7) mol/L.At L,1×10^(-6) mol/L,cardiomyocytes had no significant toxicity(P>0.05).Conclusion Shikonin has certain cytotoxicity.When the concentration of shikonin is less than 1×10^(-6) mol/L,it does not show obvious toxicity.When the concentration of shikonin is 10^(-5) mol/L,the cardiomyocytes are treated for 48h and 72h.Rat cardiomyocytes have obvious toxicity.
