摘 要:目的探讨参芪五味子颗粒(SQW)对H22肝癌荷瘤小鼠免疫抑制调节的影响。方法运用昆明种小鼠腋下荷瘤模型,称量荷瘤小鼠体重、肿瘤、脾脏、胸腺,计算抑瘤率与脏器指数。采用流式细胞术分析荷瘤小鼠脾淋巴免疫细胞荧光表达情况。ELISA法检测血清中TNF-α水平。结果参芪五味子颗粒治疗后荷瘤小鼠肿瘤缩小,对体重、胸腺指数无影响;但脾脏指数增加,脾脏中的CD3~+CD8~+阳性T细胞、CD19~+阳性B细胞、CD3~+CD4~+NK1.1~+阳性NK细胞比例以及血清中TNF-α水平显著增加(P<0.05或0.01),CD4~+CD25~+Foxp3~+阳性Treg细胞比例呈微弱的减少趋势。结论参芪五味子颗粒可增加H22荷瘤小鼠免疫器官重量,激活机体的细胞免疫和非特异性免疫,增强T细胞介导的抗肝癌免疫反应。Objective To investigate the effect of Shenqi Wuweizi granule on immunosuppressive regulation of H22 hepatoma-bearing mice. Methods Kunming mice were used to establish H22 hepatoma-bearing mice models;the weight, tumor, spleen, and thymus of H22 hepatoma-bearing mice were weighted, and the tumor inhibition rate and visceral index were calculated. The fluorescence expression of splenic lymphatic immune cells in H22 hepatoma-bearing mice was analyzed by flow cytometry, and serum TNF-α level was detected by ELISA. Results After treated with Shenqi Wuweizi granule, the tumor in H22 hepatoma-bearing mice were decreased, but there was no influence on the body weight and thymus index;spleen index was raised, and the proportions of CD3CD8positive T cells, CD19positive B cells and CD3CD4NK1.1positive NK cells in spleen and the level of TNF-α in serum were significantly increased(P<0.05 or 0.01);but the proportion of CD4CD25Foxp3positive Treg cells showed a slight decreasing trend. Conclusion Shenqi Wuweizi granule can increase the weight of immune organs in H22 hepatoma-bearing mice, activate the cellular immunity and non-specific immunity, and enhance the T cell mediated anti-hepatoma immune response.
