摘 要:以H_(3)PO_(4)-NaH_(2)PO_(4)(NaH_(2)PO_(4)浓度为39 mmol/L,pH=4.31)为运行缓冲溶液,加入体积比为2.0%正丁醇和10%乙腈为添加剂,采用20 kV分离电压,25℃柱温,214 nm检测波长,进样5 s(3.447 kPa)的仪器条件,建立了一种毛细管区带电泳同时分离分析长春花中长春碱、长春新碱、长春质碱和文多灵4种生物碱新方法。目标物分离可在14 min内完成,各目标分析物峰面积与其浓度之间线性关系良好。同时将方法应用于不同颜色的长春花茎叶和花瓣药材中这4种目标成分的分离分析,加标回收率在93.7%~108.9%之间,且相对标准偏差不大于4.1%。说明建立的新方法准确、可靠,可用于长春花药材中相关成分的质量控制。A new method of capillary zone electrophoresis was developed for simultaneous separation and determination of vinblastine,vincristine,catharanthinein and vindoline in Catharanthus roseus within 14 minutes by using H_(3)PO_(4)-NaH_(2)PO_(4)(39 mmol/L)buffer solution(pH 4.31)consisting of 2.0%(V/V)n-butanol and 10%(V/V)acetonitrile in the instrumental conditions of the separation voltage of 20 kV,column temperature of 25℃,detector wavelength of 214 nm,and injection of 5 s at 3.447 kPa.The linear relationships between peak area and concentration of each target component were good.The developed method was used to separation and analyze the four active components in the stems,leaves and petals of Catharanthus roseus with different colors.The recoveries were in the range of 93.7%-108.9%,and the relative standard deviations were less than 4.1%.The new method is accurate and reliable,and can be used for the quality control of related components in Catharanthus roseus.
