摘  要:目的 对载雌二醇介孔硅水凝胶缓释系统材料进行细胞和动物毒理学实验,为治疗宫腔粘连(Intrauterine Adhesions,IUA)新方法提供参考依据。方法 实验材料为载雌二醇介孔硅水凝胶(PHMH-Si)、载雌二醇水凝胶(PHEMA)、对照组(空白液),用死活染色法,在徕卡倒置荧光显微镜(DMi8)下观察荧光图像(活细胞和死亡细胞分别用绿色和红色标记)。同时用细胞计数试剂盒(CCK-8)检测各组HUVECs的生物活性测定细胞增殖率。并在公猪体内动物包埋进行活体实验,测量包膜厚度,了解活体组织相容性。结果 PHEMA组和PHMH-Si组的活细胞(绿色)密度变化与对照组相似,各组细胞持续增殖和铺展,培养过程中几乎未见死亡细胞。同时,PHMEA、PHMH-Si和对照组的光密度在1 d、3 d和5 d也是平坦的,结果表明PHMH-Si水凝胶具有良好的生物相容性。将载雌二醇介孔硅水凝胶(PHMH-Si)和组织相容性很好的普通水凝胶PHMEA包埋于活猪体内后取出局部组织测量包膜厚度判断炎症反应,两种水凝胶的炎症程度差异不大。结论 本组实验研究证实载雌二醇介孔硅水凝胶具有良好的组织相容性,为今后对雌激素宫内缓释系统的研究与应用提供了科学依据。Objective To conduct cell and animal toxicological experiments on materials of estradiolloaded mesoporous silica hydrogel sustained release system,and to provide a reference for the new method in the treatment of intrauterine adhesions(IUA).Methods The experimental materials were mesoporous silica hydrogel loaded with estradiol(PHMH-Si),hydrogel loaded with estradiol(PHEMA),and control group(blank liquid).The fluorescence images were observed under a Leica inverted fluorescence microscope(DMi8) after dead and alive staining(living and dead cells were marked green and red,respectively).At the same time,the bioactivity of human umbilical vein endothelial cells(HUVECs) in each group was detected by cell counting kit(CCK-8),and the in vivo experiment was carried out by animal embedment in boars.And the thickness of the capsule was measured to understand the histocompatibility in vivo.Results The change of densities of living cells(green) in PHEMA and PHMH-Si group were similar to that in the control group.Cells in each group continued proliferating and spreading,and almost no dead cells were found in the process of culture.Meanwhile,the optical densities of PHMEA,PHMH-Si and control group were flat on day 1,3 and 5,which showed that PHMH-Si had good biocompatibility.After PHMHSi and common PHMEA(with good histocompatibility) were embedded into live pigs,the local tissue was removed and the thickness of capsule was measured to determine the degree of inflammatory response.The difference in the degree of inflammation of the two hydrogels was not significant.Conclusions The experimental study showed that PHMH-Si had good histocompatibility.It’s very practical and it provides scientific basis for the research and application of the estrogen intrauterine sustained release system in the future.
