摘 要:目的:探讨Notch2在子痫前期(PE)和正常胎盘中的表达情况及对人类胎盘滋养细胞侵袭迁移功能的影响。方法:采用免疫组化检测孕早期绒毛组织、孕晚期正常胎盘和PE患者胎盘组织中Notch2蛋白的表达。免疫荧光检测滋养细胞株HTR8/SVneo的Notch2蛋白表达。Notch2 siRNA转染干扰Notch2 mRNA在HTR8/SVneo细胞株中的表达,并检测干扰Notch2表达对HTR8/SVneo细胞侵袭和迁移能力的影响。结果:免疫组化提示Notch2蛋白在早期绒毛组织滋养细胞和晚孕期正常胎盘组织的滋养层细胞中都有丰富的表达。与正常组相比,Notch2蛋白在PE患者胎盘中的表达水平降低(P<0.05)。Notch2表达下调后,HTR8/SVneo细胞的侵袭和迁移能力下降(均P<0.05)。结论:Notch2的表达水平下降导致滋养细胞的侵袭和迁移能力下降,这可能与PE的发病有重要联系。Objective:To investigate the expression of Notch2 in the preeclampsia(PE)and normal placenta,and explore its effect on the invasion and migration of human placental trophoblast cells.Methods:Immunohistochemistry was used to detect the expression of Notch2 protein in villi tissue,normal maternal placenta and placenta of PE patients.The expression of Notch2 protein in trophoblast cell line HTR8/SVneo was detected by immunofluorescence assay.HTR8/SVneo cell lines were transfected with Notch2 siRNA to attenuate the expression of Notch2 mRNA.The effects of Notch2 knocked down on invasion and migration ability of HTR8/SVneo cells were detected.Results:Immunohistochemistry indicated that Notch2 protein was highly expressed in trophoblast cells of early villi tissue and last trimester placenta tissue.Compared with the normal group,the expression level of Notch2 protein in the placenta of PE patients was decreased(P<0.05).Down-regulation of Notch2 inhibited invasion and migration ability of HTR8/SVneo cells(both P<0.05).Conclusion:Down-regulation of Notch2 could decrease the invasion and migration ability of trophoblast cells,which may be related to the pathogenesis of PE.
