摘  要:目的 探讨帕瑞昔布钠对BT474乳腺癌细胞增殖、迁移与侵袭能力的影响,及其作用机制。方法 体外培养BT474乳腺癌细胞,分为对照组和低、中、高剂量实验组。对照组在正常培养基中培养24 h;低、中、高剂量实验组分别置于含100,300和500μmol·L^(-1)帕瑞昔布钠的RPMI-1640培养基中培养24 h。用细胞计数法-8(CCK-8)实验检测细胞的增殖能力,用划痕实验检测BT474细胞的迁移能力,用Transwell实验检测细胞的侵袭能力,用Western blot法检测细胞中B淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)、Bcl-2相关X蛋白(Bcl-2-associated X protein,Bax)和胱天蛋白酶-3(cysteinyl aspartate specific proteinase-3,Caspase-3)的表达情况。结果 处理24 h后,低、高剂量实验组和对照组的细胞增殖抑制率分别为(20.28±1.33)%,(67.91±2.35)%和(100.00±0)%,细胞迁移率分别为(20.84±2.15)%,(5.77±0.72)%和(26.03±2.09)%,细胞侵袭数分别为(1.72×10~4±397.00),(0.83×10~4±532.00)和(2.04×10~4±925.00)个,Bcl-2蛋白相对表达量分别为0.92±0.04,0.17±0.04和1.79±0.11,Bax蛋白相对表达量分别为0.51±0.05,1.21±0.11和0.38±0.02,Caspase-3蛋白相对表达量分别为0.82±0.08,1.98±0.17和0.33±0.06;低、高剂量实验组的上述指标与对照组比较,差异均有统计学意义(均P<0.05)。结论 帕瑞昔布钠可抑制BT474乳腺癌细胞增殖、迁移和侵袭的能力,其机制可能与调控细胞凋亡信号通路有关。Objective To investigate the effect of parecoxib sodium on the proliferation,migration and invasion of BT474 breast cancer cells and its mechanism.Methods BT474 breast cancer cells were cultured in vitro and divided into control and experimental-L,-M,-H groups.The control group was cultured for 24 h in normal medium and the experimental-L,-M,-H groups were grown in RPMI-1640 medium containing 100,300,and 500 μmol·L^(-1) parecoxib sodium for 24 h,respectively.Cell counting kit-8(CCK-8) test was used to detect the proliferation ability of cells,scratch healing test was used to detect the migration ability of cells,Transwell test was used to detect the invasiveness of cells,and Western blot was used to detect the expression levels of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax),and cysteinyl aspartate specific proteinase-3(Caspase-3) proteins in BT474 breast cancer cells.Results After 24 h of treatment,the rates of cell proliferation inhibition in the experimental-L,-H groups and control group were( 20.28 ± 1.33) %,( 67.91 ± 2.35) % and( 100.00 ± 0) %;the rates of cell mobility were( 20.84 ± 2.15) %,( 5.77 ± 0.72) % and( 26.03 ± 2.09) %;the number of cell invasion were( 1.72 × 10~4± 397.00),( 0.83 × 10~4± 532.00) and( 2.04 × 10~4± 925.00);Bcl-2 protein expression were 0.92 ± 0.04,0.17 ± 0.04 and 1.79 ± 0.11;Bax protein expression were 0.51 ± 0.05,1.21 ± 0.11and 0.38 ± 0.02;Caspase-3 protein expression were 0.82 ± 0.08,1.98 ± 0.17 and 0.33 ± 0.06.Compared with the control group,the experimental-L,-H groups showed statistically significant differences in the above indexes( all P < 0.05).Conclusion Parecoxib sodium can inhibit the proliferation,migration and invasion of BT474 breast cancer cells,which mechanism may be associated with regulating the signaling pathways of apoptosis.
