摘  要:克隆猪OASL2基因并对其进行序列分析。采用PCR技术扩增猪OASL2基因,连接至pCE2 TA/Blunt-Zero载体并将其转化至DH5α化学感受态细胞,测序获得猪OASL2基因CDS全长,对其进行生物学功能预测以及构建系统进化树。结果显示,猪OASL2基因全长为633 bp,编码211个氨基酸,蛋白分子质量为23.94 ku,理论等电点为8.16。与猪OASL1相似性为98.1%,氨基酸相似性为97.6%,与其它物种OASL相似性为52.3%~72.0%,氨基酸相似性为9.8%~58.1%。系统进化树表明猪OASL2与猪OASL1位于同一分支,与牛OASL位于同一亚分支,亲缘关系较近。氨基酸序列比对结果表明猪OASL2包含P-loop、D-box以及保守位点LLRL。The experiment was to clone porcine OASL2 gene and perform sequence analysis.The porcine OASL2 gene was amplified by PCR,ligated into pCE2 TA/Blunt-Zero vector,and transformed into DH5αchemoreceptor cells.The full-length CDS of porcine OASL2 gene was obtained by sequencing.Prediction of biological function of it and construction of phylogenetic tree.The results showed that the full length of porcine OASL2 gene was 633 bp,encoding 211 amino acids,with a molecular weight of 23.94 ku and a theoretical isoelectric point of 8.160.The nucleotide similarity between porcine OASL2 and porcine OASL1 was 98.1%,and the amino acid similarity was 97.6%.The nucleotide similarity between porcine OASL2 and other species was 52.3%~72.0%,and the amino acid similarity was 9.8%~58.1%.The phylogenetic tree showed that porcine OASL2 and porcine OASL1 were located in the same branch,and bovine OASL were located in the same subbranch,with close genetic relationship.Amino acid sequence alignment results showed that porcine OASL2 contained P-loop,D-box and conserved site LLRL.
