高效液相色谱法同时测定减肥类保健食品中6种蒽醌苷元的含量

摘　　要：目的建立高效液相色谱法(HPLC)同时测定减肥类保健食品中橙黄决明素、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚6种蒽醌苷元含量的方法。方法减肥类保健食品以甲醇沸水浴提取、稀盐酸超声水解、二氯甲烷萃取等前处理后,采用BDS HYPERSIL C_(18)色谱柱分离,以甲醇-0.1%磷酸为流动相进行梯度洗脱,经二极管阵列检测器分析,外标法定量。结果6种蒽醌苷元在1~50μg/ml质量浓度范围内线性良好,r均高于0.998;检出限均为0.04 mg/g。平均加标回收率为85.2%~98.1%;精密度(n=6)在7.0%以内。结论该方法可用于减肥类保健食品中6种蒽醌苷元的同时检测。Objective To establish an HPLC method for the simultaneous determination of 6 anthraquinone aglycones(aurantio-obtusin,aloe-emodin,rhein,emodin,chrysophanol and physcion)in weight-reducing health food.Methods After extraction with methanol in boiling water bath,the extract was treated by supersonic extraction with diluted hydrochloric acid,and extracted with dichloromethane.The sample was separated with a BDS HYPERSIL C_(18) chromatographic column,using methanol and 0.1%phosphoric acid as the mobile phase,analyzed using a diode array detector and quantified with reference to an external standard.Results The six anthraquinone aglycones had good linearities in the mass concentration range of 1-50μg/ml,with the correlation coefficients greater than 0.998.The detection limits of aurantio-obtusin,aloe-emodin,rhein,emodin,chrysophanol and physcion were all 0.04 mg/g.The average recoveries were 85.2%-98.1%and the relative standard deviations were less than 7.0%(n=6).Conclusion The established method can be used for the simultaneous determination of 6 anthraquinone aglycones in health weight-reducing food.
