摘 要:热碱预处理对于污泥蛋白质的溶出具有显著的促进作用。本研究以工业热碱工艺获得的粗污泥混合液作为原料,以蛋白质、胞外聚合物和胞内物质含量为指标,探究蛋白酶协同超声与DNA提取酚试剂水解对污泥蛋白质的提取以及自身脱水稳定性的促进效果。结果表明:最佳的蛋白酶种类和初始酶加量分别为碱性蛋白酶和4000U/g,此时滤液蛋白质含量为883.43mg/L,较于未加入蛋白酶组提高23.98%。协同处理后,污泥混合液的S-EPS含量升高83.44%,LB-EPS和TB-EPS分别降低33.11%和24.95%,同时SEPS中蛋白质与多糖的比值提高了100.40%,污泥中疏松附着型、紧密附着型胞外聚合物以及胞内物质含量显著降低,证明协同处理能进一步提升污泥混合液的稳定性。Thermal alkali pretreatment can significantly promote the dissolution of sludge protein.In this study,the sludge mixture raw material obtained from thermal alkali pretreatment process,was used for the investigation of protein enhanced dissolution.Wherein,the contents of protein,extracellular polymer and intracellular substances were used as indicators to explore the promoting effects of the combination of protease synergistic ultrasound and DNA extraction phenol reagent on the raw material’s hydrolysis and dehydration stability.Results showed that the optimal protease addition were determined.Results showed that with the 4000 U/g alkaline protease addition,the protein content of the filtrate was 883.43 mg/L,which was 23.98% higher than that of the group without protease.After synergistic treatment,the S-EPS concentration increased by 83.44%,while the LB-EPS and TB-EPS decreased by 33.11% and 24.95% respectively.At the same time,the ratio of protein to polysaccharide in SEPS increased by 100.40%,indicating the contents of loosely and tightly attached extracellular polymers or intracellular substances in sludge decreased significantly.All the results confirmed that synergistic treatment could further improve the dewatering performance and stability of sludge.
