伯氏疟原虫Pb22截短蛋白免疫血清的传播阻断能力的研究

摘  要:为探讨伯氏疟原虫(Plasmodium berghei)Pb22蛋白免疫血清的传播阻断能力,应用原核表达系统高效表达Pb22截短蛋白免疫小鼠后获得免疫血清。IFA实验证明Pb22截短蛋白免疫血清均可与疟原虫天然抗原结合。通过传播阻断实验,比较了Pb22截短蛋白和全长蛋白免疫血清的传播阻断效果,证明截短蛋白和全长蛋白对雄配子体出丝均有抑制作用,结果表明全长蛋白免疫小鼠的雄配子体出丝与对照组相比显著下降,截短蛋白免疫小鼠的雄性配子体出丝具有下降趋势但无统计学差异。全长蛋白和截短蛋白免疫小鼠的动合子形成数量较对照组均显著下降。以上结果表明抗Pb22截短蛋白免疫血清具有传播阻断能力,但阻断效果不如全长蛋白免疫血清。In order to investigate the transmission blocking activity of the Plasmodium berghei Pb22 protein immunized sera,we expressed Pb22 truncated protein by using prokaryotic expression system and obtained antisera after immunized with mice.IFA test showed that the immune serum of Pb22 truncated protein could bind to the natural antigen of Plasmodium.Through the transmission blocking experiment,the transmission blocking activity of the Pb22 truncated protein and the full-length protein immunized sera were compared.It was proved that the truncated protein and the full-length protein both had an inhibitory effect on exflagellation of male gametocytes.The number of exflagellation centers of male gametocytes from full-length protein immunized mice decreased significantly when compared with control group.The exflagellation of male gametocytes of truncated protein immunized mice showed a downward trend,but there was no statistical difference.The number of ookinete formed in mice immunized with full-length protein and truncated protein was significantly lower than that in the control group.These results indicate that the anti-Pb22 truncated protein immune sera have the ability to block transmission,but the blocking effect is not as good as the full-length protein immune sera.
