摘 要:目的观察散结消瘿颗粒对实验性自身免疫性甲状腺炎(EAT)大鼠超氧化物歧化酶(SCD)、丙二醛(MDA)以及NADPH氧化酶亚基p22phox、gp91phox基因和蛋白表达的影响,评价其对氧化应激的改善和治疗作用。方法建立EAT大鼠模型,设立正常组、模型组、散结消瘿颗粒高(SJ XY 39.2 g·kg·d^(-1))/低(SJXY 4.9 g·kg·d^(-1))剂量组采用酶联免疫吸附法测定血清甲状腺球蛋白抗体(TgAb)、甲状腺过氧化物酶抗体(TPOAb)水平和SOD、MDA含量,采用RT-PCR和Western-Blot检测甲状腺p22phox、gp91phox基因和蛋白表达。结果与正常组相比,模型组TgAb、TPOAb水平、p22phox、gp91phox基因和蛋白表达均升高(均P<0.01)SOD/MDA水平降低(P<0.01)。与模型组相比,SJXY高、低剂量组TgAb、TPOAb水平和p22phox、gp91phox基因和蛋白表达均降低(均P<0.05),SJXY高剂量组SOD/MDA水平升高(P<0.05)。与低剂量组相比,SJXY高剂量组TgAb、TPOAb、p22phox、gp91phox基因和蛋白表达均呈下降趋势,SOD/MDA水平呈上升趋势。结论散结消瘿颗粒具有显著降低EAT模型大鼠氧化应激损伤的作用,其机制可能是通过下调甲状腺组织NADPH氧化酶亚基p22phox、gp91phox基因和蛋白表达,调节SOD/MDA水平平衡,同时降低TgAb、TPOAb水平来实现,且试验显示高剂量优于低剂量。Objective To observe the effects of Sanjie Xiaoying granule on the expression of superoxide dismutase(SOD),malondialdehyde(MDA)and NADPH oxidase subunits p22 phox and gp91 phox in experimental autoimmune thyroiditis(EAT)rats,and to evaluate the improvement and therapeutic effect of Sanjie Xiaoying granule on oxidative stress.Methods The normal group,model group,high-dose(39.2 g·kg·d^(-1))and low-dose(4.9 g·kg·d^(-1))groups of Sanjie Xiaoying granule were established.The levels of TgAb and TPOAb and the contents of SOD and MDA in serum of rats were determined by ELISA.The gene and protein expressions of NADPH oxidase subunit p22 phox and gp91 phox in thyroid of rats were detected by RT-PCR and Western-blot.Results Compared with normal group,the levels of TgAb,TPOAb and the gene and protein expre ssions of p22 phox and gp91 phox in model group were significantly increased(all P<0.01),while the level of SOD/MDA was significantly decreased(P<0.01).Compared with model group,the levels of TgAb and TPOAb and the gene and protein expressions of p22 phox and gp91 phox in high-dose and low-dose groups were significantly decreased(all P<0.05),while the level of SOD/MDA in high-dose group was significantly increased(P<0.05).Compared with the low-dose group,the levels of TgAb and TPOAb and the gene and protein expressions of p22 phox and gp91 phox in high-dose group showed a downward trend,while the level of SOD/MDA showed an upward trend.Conclusions Sanjie Xiaoying granule could significantly reduce oxidative stress injury in EAT model rats.The mechanism might be through down-regulating the gene and protein expression of NADPH oxidase subunit p22 phox and gp91 phox in thyroid tissues,regulating the balance of SOD/MDA level,and reducing the levels of TgAb and TPOAb,and the test showed that high dose wa s better than the low dose.
