猪圆环病毒2型亚单位疫苗的研制及其ELISA抗体评价

摘　　要：为制备并筛选出安全性更好、免疫效果更佳的猪圆环病毒2型(PCV-2)亚单位疫苗,本研究选取本地流行毒株PCV2 TX01株,通过杆状病毒表达系统对Cap蛋白进行高效表达,采用水性佐剂制备成灭活疫苗,从临床症状、日增重和ELISA抗体水平等方面评价免疫后对仔猪的安全性和血清学效力。结果表明,疫苗免疫后仔猪精神、食欲和行为活动均正常,接种部位未见红肿等不良症状,证实疫苗对仔猪安全。免疫组仔猪ELISA抗体效价在1:400~1:800,说明该疫苗免疫后可以有效地刺激仔猪产生体液免疫,可以作为预防猪圆环病毒相关疾病的候选疫苗。To prepare and screen porcine circovirus type 2(PCV-2) subunit vaccine with better safety and immune effect. In this study, the local epidemic strain PCV2 TX01 was selected, and Cap protein was highly expressed by baculovirus expression system, and inactivated vaccine was prepared by water-based adjuvant. The safety and serological efficacy of immunization on piglets were evaluated from clinical symptoms, daily gain, ELISA antibody level and other aspects. The results showed that the spirit, appetite and behavior of piglets were normal after immunization with the vaccine, and no adverse symptoms such as redness and swelling were observed at the vaccination site, which confirmed that the vaccine was safe for piglets. The titer of ELISA antibody of piglets in immunization group was 1:400 ~ 1:800, indicating that the vaccine could effectively stimulate the humoral immunity of piglets after immunization, and could be used as a candidate vaccine to prevent porcine circovirus-related diseases.
