摘 要:本实验研究了恩诺沙星乳在异育银鲫(Carassius auratus gibelio)和斑点叉尾鮰(Ictalurus punetaus)体内的药代动力学规律。异育银鲫和斑点叉尾鮰单剂量口灌恩诺沙星乳(10 mg/kg bw),采用高效液相色谱-串联质谱(HPLC-MS/MS)法测定了给药后的血药浓度。结果显示,恩诺沙星乳在异育银鲫和斑点叉尾鮰体内的达峰浓度(C_(max))分别为4.66μg/mL和3.95μg/mL,相应的达峰时间(T_(max))分别为6 h和8 h;消除半衰期(T_(1/2z))分别为20.05 h和22.06 h;药时曲线下面积(AUC 0-∞)分别为228.26μg·h/mL和189.55μg·h/mL。其主要代谢产物环丙沙星在异育银鲫和斑点叉尾鮰体内的C_(max)分别为0.053μg/mL和0.043μg/mL,相应的T_(max)均为12 h;T_(1/2z)分别为45.79 h和31.66 h;AUC_(0-∞)分别为5.24μg·h/mL和3.40μg·h/mL。恩诺沙星乳在异育银鲫和斑点叉尾鮰体内的生物转化率分别为2.07%和1.74%。单剂量的恩诺沙星乳(10 mg/kg bw)适用于恩诺沙星对异育银鲫和斑点叉尾鮰病原菌的最小抑菌浓度(MIC)小于0.58μg/mL和0.49μg/mL的敏感菌。The concentrations of enrofloxacin and ciprofloxacin were determined by high performance liquid chromatography tandem mass spectrometry(HPLC-MS/MS)at different time points after a single oral administration of enrofloxacin emulsion at 10 mg/kg body weight to explore the pharmacokinetics of enrofloxacin emulsion in Carassius auratus gibelio and Ictalurus punetaus.The maximum plasma concentration(C_(max))of enrofloxacin in C.auratus gibelio and I.punetaus was 4.66μg/mL and 3.95μg/mL,respectively.The time to peak concentration(T_(max))was 6 h and 8 h,respectively.The elimination half-lives(T_(1/2z))were 20.05 h and 22.06 h,respectively.The areas under the drug-time curve(AUC_(0-∞))were 228.26μg·h/mL and 189.55μg·h/mL,respectively.As for its main metabolite ciprofloxacin,C_(max) was 0.053μg/mL and 0.043μg/mL,respectively,both of T_(max) were 12 h.T_(1/2 z) were 45.79 h and 31.66 h,respectively.The AUC_(0-∞) were 5.24μg·h/mL and 3.40μ·hg//mL,respectively.The AUC ratios of ciprofloxacin and enrofloxacin in C.auratus gibelio and I.punetaus plasma were 2.07%and 1.74%,respectively.Enrofloxacin emulsion administration was suitable for enrofloxacin with a minimum inhibitory concentration(MIC)of less than 0.58μg/mL and 0.49μg/mL to pathogenic bacteria of C.auratus gibelio and I.punetaus.
