摘 要:[目的]探索miR-429在雪旺细胞中通过调节P13K/AKT信号通路在高糖诱导的雪旺细胞损伤中发挥保护作用的能力。[方法]细胞凋亡检测和CCK-8法评估大鼠雪旺细胞系RSC96在高糖培养条件下的细胞损伤情况,过表达或敲低miR-429转染细胞后,WB检测p/t-P13K和p/t-AKT蛋白相对表达情况,细胞凋亡检测和CCK-8法评估雪旺细胞损伤情况。[结果]高糖培养诱发雪旺细胞凋亡率升高(P<0.001)和细胞增殖能力降低(P<0.001),p/t-P13K和p/t-AKT显著降低(P<0.05);过表达miR-429后p/t-P13K和p/t-AKT显著提升(P<0.05),细胞凋亡与坏死比率显著下调(P<0.05),细胞增殖能力显著提升(P<0.05);敲低miR-429后,p/t-P13K和p/t-AKT显著下降(P<0.05),细胞凋亡与坏死比率显著上调(P<0.05),细胞增殖能力显著下降(P<0.05)。[结论]miR-429的过表达能够抑制雪旺细胞由于高糖培养导致的p/t-P13K和p/t-AKT下调(P<0.05),从而显著下调雪旺细胞由于高糖诱导的细胞凋亡(P<0.05),并提高雪旺细胞在高糖培养状态下的细胞增殖率(P<0.05)。综上,miR-429能够通过调节P13K/AKT信号通路在高糖诱导的雪旺细胞损伤中发挥保护作用。[Objective]To explore the ability of miR-429 in Schwann cells to play a protective role in high glucose-induced Schwann cell injury by regulating the P13 K/AKT signaling pathway.[Method]Apoptosis detection and CCK-8 assay were used to evaluate the cell injury of rat Schwann cell line RSC96 under high glucose culture conditions.After overexpression or knockdown of miR-429 transfected cells,Western Blotting detects p/t-P13 K and p/t-AKT protein relative expression,apoptosis detection and CCK-8 assay to evaluate Schwann cell injury.[Result]High glucose culture induced increased Schwann cell apoptosis rate(P<0.001)and decreased cell proliferation ability(P<0.001),p/T-P13 K and P/T-Akt were significantly decreased(P<0.05);After overexpression of miR-429,p/t-P13 K and p/t-AKT were significantly increased(P<0.05),the ratio of apoptosis and necrosis was significantly down-regulated(P<0.05),and the cell proliferation ability was significantly improved(P<0.05);After knocking down miR-429,p/t-P13 K and p/t-AKT decreased significantly(P<0.05),the ratio of apoptosis and necrosis was significantly increased(P<0.05),and the cell proliferation ability was significantly decreased(P<0.05).[Conclusion]Overexpression of miR-429 can inhibit the down-regulation of p/t-P13 K and p/t-AKT in Schwann cells due to high glucose culture(P<0.05),thereby significantly down-regulating the apoptosis of Schwann cells due to high glucose(P<0.05),and increase the cell proliferation rate of Schwann cells under high glucose culture(P<0.05).In summary,miR-429 can play a protective role in Schwann cell injury induced by high glucose by regulating the P13 K/AKT signaling pathway.
