摘  要:[目的]探讨催产素对慢性应激后抑郁大鼠的行为影响及分子机制。[方法]大鼠分对照、实验组(模型、治疗组),实验组以慢性应激构建抑郁模型,治疗组于造模29d腹腔注射催产素,35 d检测行为学;ELISA检测催产素、皮质酮;Western Blotting检测纹状体催产素受体、多巴胺转运体及海马C-Fos;qPCR检测纹状体炎症和趋化因子。[结果]较模型组,治疗组体重增加量(13.29±3.52 g)、旷场试验得分(48.17±6.33)及糖水消耗(61.34%±8.43%)升高;催产素(238.41±21.53 pg/mL)、皮质酮(28.19±3.62 pg/mL)水平恢复;催产素受体与多巴胺转运体增加,C-Fos降低;IL6、IL8、TNF-α、CXCL2、CXCL10等因子降低,差异有统计学意义(P<0.05)。[结论]催产素对抑郁模型行为具有治疗效果,治疗后催产素、催产素受体与多巴胺转运体水平提高,皮质酮、炎症和趋化因子水平降低。[Objective]To investigate the effect of oxytocin on behavior and molecular mechanism of chronic stress depression rats.[Method]Rats were divided into control group and experimental group(model and treatment group).Chronic stress was used to construct depression model in experimental group.ELISA was used to detect oxytocin and corticosterone.The striatum oxytocin receptor,dopamine transporter and hippocampal C-Fos were detected by Western Blotting.Striatum inflammation and chemokines were detected by qPCR.[Result]Compared with model group,the differences in the treatment group were as follows.Body weight gain(13.29±3.52 g),open field test score(48.17±6.33)and sugar water consumption(61.34%±8.43%)were higher.The levels of oxytocin(238.41±21.53 pg/mL)and corticosterone(28.19±3.62 pg/mL)recovered.Oxytocin receptor and dopamine transporter increased,C-Fos decreased;IL6,IL8,TNF-α,CXCL2,CXCL10 were decreased.The differences mentioned above were statistically significant(P<0.05).[Conclusion]Oxytocin has therapeutic effect on the behavior of depressive model.After treatment,the levels of oxytocin,oxytocin receptor and dopamine transporter are increased,while the levels of corticosterone,inflammation and chemokine are decreased.
