摘  要:稀土金属冶选尾矿库渗漏导致地下水污染会严重威胁当地人民的生命安全,ICR小鼠是当今评估人类疾病健康的一个最佳模式动物。为评估稀土金属冶选尾矿库渗透导致地下水污染对生物的遗传损伤效应以及可能的分子机制,首先调查稀土金属冶选尾矿库周边地下水的核心污染成分;其次分析了受污染的地下水对ICR小鼠在肝脏的遗传损伤效应,进一步利用转录组数据深入研究污染因子对ICR小鼠的遗传损伤的可能分子机制。结果表明,矿区的核心污染成分是盐离子和氨氮等,尾矿库污染地下水导致ICR小鼠肝体比先升后降,组织内部发现铁血红素沉积、细胞损伤及组织间隙增大等现象,细胞水平上发现线粒体结构异常且出现自噬溶酶体,单细胞凝胶电泳结果显示DNA损伤率增高。对转录组测序数据的分析表明,发现135个上调基因,95个下调基因;KEGG通路分析显示差异基因主要富集在MAPK信号通路、AMPK信号通路、内质网中的蛋白质加工、FoxO信号通路、以及多种癌变通路等方面。研究结果为揭示尾矿库周边地下水污染对生物体的遗传损伤机制提供理论基础。Groundwater contamination due to leakage from rare earth metal metallurgy tailing ponds would seriously threaten the life safety of local people,and ICR mice is today one of the best model animals for evaluating the health of human diseases.In order to evaluate the genetic damage effect of groundwater contamination to organisms as a result of rare-earth metal metallurgical ore infiltration,and the possible molecular mechanism,this project first investigated the core pollution components of the groundwater surrounding the rare-earth metal metallurgy ore,secondly analyzed the effect of genetic damage to the liver of ICR mice from contaminated groundwater;The transcriptome data were further utilized to deeply investigate the possible molecular mechanisms of the genetic damage of ICR mice by pollution factors.The results showed that the core pollution components in the mining area are salt ions and ammonia nitrogen and so on;The tailing pond contaminated the groundwater to cause ICR mice liver body ratio to rise first and then fall;Hemosiderin deposition,cell damage and increased interstitial space were found in the interior of the tissue;Abnormal mitochondrial structure and the appearance of autophagolysosomes were found at the cellular level,and the results of single-cell gel electrophoresis showed an increased rate of DNA damage;Analysis of the transcriptome sequencing data showed that 135 upregulated genes,95 downregulated genes were found;KEGG pathway analysis showed that the differential genes were mainly enriched in the MAPK signaling pathway,AMPK signaling pathway,protein processing in endoplasmic reticulum,FOXO signaling pathway,and multiple pathways of carcinogenesis.This study provided a theoretical basis for uncovering the mechanism of genetic damage to organisms from groundwater pollution at the periphery of a tailing pond.
