摘 要:目的 基于网络药理学探讨当归补血汤治疗放射性心肌病的物质基础和机制。方法 通过检索中医药综合数据库筛选黄芪、当归的潜在活性成分,构建药物与疾病的交集靶点,采用STRING数据库构建靶点蛋白质-蛋白质相互作用(PPI)网络,对交集靶点进行基因本体论(GO)功能分析和基于京都基因和基因组百科全书(KEGG)通路富集分析。结果 筛选出潜在活性成分29个、药物与疾病的交集靶点82个、21个PPI核心基因。GO功能分析显示交集靶点主要参与氧化应激反应、细胞凋亡、细胞因子活性等生物过程。KEGG通路富集分析显示当归补血汤主要参与IL-17信号通路、PI3K/Akt通路、NF-κB信号通路、 JAK-STAT信号通路、AGE-RAGE信号通路等。结论 网络药理学研究显示当归补血汤治疗放射性心肌病的作用机制主要为抗氧化损伤、调节炎症反应、调节细胞凋亡等。Objective To explore the material basis and mechanism of Danggui Buxue Tang in the treatment of radiation induced heart disease based on network pharmacology. Methods The potential active components of Astragalus membranaceus and Angelica sinensis were screened by searching the comprehensive database of traditional Chinese medicine, and the intersection targets of drugs and diseases were constructed. The protein-protein interaction(PPI) network was constructed by using STRING database, and gene ontology(GO) function analysis and KEGG pathway enrichment analysis were performed on the intersection targets.Results 29 potential active ingredients, 82 intersection targets of drugs and diseases, and 21 PPI core genes were screened out. GO enrichment indicates that intersection targets are mainly involved in response to reactive oxygen species, apoptotic signaling pathway, sytokine activity and other biological processes. KEGG pathway enrichment indicated Danggui Buxue Tang was mainly involved in IL-17 signaling pathway,PI3K/Akt pathway, NF-κB signaling pathway, JAK-STAT signaling pathway, AGE-RAGE signaling pathway.Conclusion The network pharmacology study showed that the mechanism of Danggui Buxue Tang in the treatment of radiation induced heart disease mainly anti-oxidative damage, regulating inflammatory response, regulating apoptosis and so on.
