摘 要:为明确猪脾转移因子(TF)的鸡临床安全性和有效性,采用自制的3批猪脾TF,分别以0.1和1.0 mL剂量肌内注射1、14日龄肉种鸡和1、7、14、21日龄肉鸡,以0.2和1.0 mL剂量肌内注射8、26周龄肉种鸡,并设立不注射对照组,共有9800羽鸡用于临床安全性试验;将3批猪脾TF以0.1 mL剂量分别与1.0羽份新城疫病毒(NDV)活疫苗(La Sota株)联合免疫1、14日龄肉种鸡和1、7、14、21日龄肉鸡,以0.2 mL剂量分别与1.0羽份NDV活疫苗(La Sota株)联合免疫8、26周龄肉种鸡,并设立单独免疫对照组,共有6078羽鸡用于临床有效性试验,采用血凝抑制(HI)试验和淋巴细胞增殖试验,分别测定NDV HI抗体和淋巴细胞刺激指数(SI)。安全性试验结果显示,鸡群肌内注射TF后无应激反应,其精神状况、采食、饮水、粪便、注射部位反应情况均正常,剖检均未发现组织、器官病变,死淘率不高于对照组鸡群,肉种鸡产蛋情况正常;有效性试验结果显示,免疫后14、21 d联合免疫组NDV HI抗体滴度和淋巴细胞SI分别比单独免疫组提高了1.1~1.7、0.243~0.632,差异均显著(P<0.05)。结果表明,猪脾TF对临床肉种鸡群和肉鸡群安全,并且具有增强临床鸡群免疫功能的作用。本研究为TF的临床应用提供了数据支持。In order to determine the clinical safety and efficacy of porcine spleen transfer factor(TF)in chickens,three batches of self-made porcine spleen TF were intramuscularly injected at the doses of 0.1 and 1.0 mL respectively into broiler breeders at the ages of 1 day and 14 days and broilers at the ages of 1,7,14 and 21 days,at the doses of0.2 and 1.0 mL respectively into broiler breeders at the ages of 8 and 26 weeks,and a control group without injection was prepared,a total of 9 800 chickens were used in the clinical safety test. Three batches of porcine spleen TF were co-immunized with 1.0 attenuated chicken Newcastle disease virus(NDV)(La Sota strain)at a dose of 0.1 mL in the broiler breeders at the ages of 1 and 14 days and broilers at the ages of 1,7,14 and 21 days,respectively,and at a dose of 0.2 mL in broiler breeders at the ages of 8 and 26 weeks,and a control group with single injection was prepared,a total of 6 078 chickens were used in the clinical efficacy test. NDV hemagglutination inhibition(HI)antibody and lymphocyte stimulation index(SI)were measured by HI test and lymphocyte proliferation test,respectively. It was shown that,by the results of safety test,no stress reaction was observed in chickens after intramuscular injection of TF,their mental status,feed intake,water drinking,feces and reaction at the injection parts were normal,no tissue and organ lesions were found during the autopsy,the mortality rate was lower than that of the control group,and the laying of broilers was normal;by the results of effectiveness test,the NDV HI antibody titer and lymphocyte SI of the group with combined immunization were from 1.1 to 1.7 and from 0.243 to 0.632 higher than those of the one with single immunization,respectively,14 and 21 days after immunization,with significant difference(P< 0.05). In conclusion,porcine spleen TF was safe and could enhance the immune function of clinical broiler breeders and broiler chickens. A reference was thus provided for future clinical application of TF.
