摘  要:【目的】分析贵州境内月鳢自然群体间的遗传关系及其遗传背景,为贵州土著鱼类资源的保护和开发利用提供理论依据。【方法】以贵州境内长江流域(乌江、清水江)和珠江流域(都柳江、猫营河)的4个月鳢群体(共130尾月鳢样本)为研究对象,采用PCR技术扩增月鳢线粒体Cyt b编码区,分析月鳢自然群体的遗传结构。【结果】贵州月鳢Cyt b基因946 bp序列的A+T含量为53.7%,高于G+C含量(46.3%),其碱基组成表现为C的含量最高、G的含量最低,呈较强的反G偏倚性;130个月鳢样本定义6种单倍型,总体核苷酸多样性指数为0.00198,单倍型多样性指数为0.734;群体间平均遗传距离为0.00229,猫营河-都柳江群体的遗传距离最大(0.00308),乌江-猫营河群体的最小(0.00131)。【结论】贵州月鳢整体遗传多样性水平低,群体间存在一定的遗传分化。【Objective】The genetic relation and background of Channa argus population in Guizhou are analyzed,which provide theory basis for the protection and utilization of local fish resource in Guizhou.【Method】Taking the C.argus population with four-month-age(a total of 130 C.argus samples)from Yangtze River basin(Wu River,Qingshui River)and Pearl River basin(Duliu River,Maoying River)as the study object,then the Cyt b coding area of mitochondrion of C.argus is amplificated by PCR technology,so as to analyze the genetic structure of C.argus population.【Result】The A+T content in 946 bq sequence of Cyt b gene of Guizhou C.argus is 53.7%,which is higher than the G+C content(46.3%),and C content is the highest,while G content is the lowest which shows a strong trend of resistant to G in the base composition.130 C.aryus samples determines 6 haplotypes,and the total diversity index of nucleotide is 0.00198,while the diversity index of haplotype is 0.734.The average genetic distance in population is 0.00229,among which the genetic distance of C.argus population from Maoying River-Duliu River is the biggest(0.00308),and that of Wu River-Maoying River is the smallest(0.00131).【Conclusion】The level of genetic diversity of C.argus population in Guizhou is low in general,and there are genetic differentiation among populations to some degree.
