摘  要:甘蔗褐条病和梢腐病是为害甘蔗叶部的灾害性真菌病害。为了改善马铃薯葡萄糖琼脂(PDA)培养基存在病菌生长速度缓慢、产孢时间长、产孢数量少等问题,对甘蔗褐条病与梢腐病病原菌快速大量产孢的培养方法进行了研究。结果表明甘蔗褐条病菌接种于褐条病菌产孢培养基后,经去除菌丝处理后暗培养7 d可产生大量褐条病菌分生孢子;甘蔗梢腐病接种于梢腐病菌产孢培养基后,振荡培养3 d可产生大量分生孢子。本方法培养基原料简单易得,成本低廉,步骤简单,操作方便,污染率低,与PDA培养基传统培养方法相比,培养周期显著缩短,每个视野分生孢子增加6倍以上,充分满足了甘蔗褐条病和梢腐病深入研究与防控的需求,为甘蔗褐条病和梢腐病病菌变异动态、致病力差异、筛选抗病品种及绿色高效杀菌剂等应用研究提供了基础条件及技术支撑。Sugarcane pokkah boeng and brown stripe are the disastrous fungal disease that harms the leaves of sugarcane.In order to improve the problems of slow growth rate,long spore production time and low number of spores,the culture methods for rapid and large-scale sporulation of sugarcane pokkah boeng and brown stripe pathogen were studied.The results showed that after inoculation in spore-producing medium of sugarcane brown stripe,a large number of conidia of sugarcane brown stripe had produced when dark culture for 7 days after removed mycelium.A large number of conidia of sugarcane pokkah boeng had produced after shaking culture for 3 days in spore-producing medium of sugarcane pokkah boeng.The sporulation technology has merits including simple medium components,easily available raw materials,low raw material cost,convenient operation and low contamination rate.Compared with the PDA cultural method,the incubation period was significantly shortened,and the conidia per field increased by more than six times,which met the needs of in-depth study and control of sugarcane pokkah boeng and brown stripe.The results provided basic conditions and technical support for the application of sugarcane pokkah boeng and brown stripe pathogen variation dynamics,pathogenicity difference,screening of resistant varieties and high-efficiency green fungicides.
