甘薯块根响应长喙壳菌(Ceratocystis fimbriata)侵染的转录组测序分析

摘  要:以不侵染黑斑病菌长喙壳菌(Ceratocystis fimbriata Ellis et Halsted)的抗病甘薯品种南京92和感病品种徐薯18的薯块为对照组,以侵染时间1、3、7 d时的薯块为处理组进行转录组测序。测序结果表明:在抗病品种中找到36127个差异表达基因(differential expressed genes,DEGs),其中上调、下调表达的基因分别有17387、18740个;在感病品种中找到44663个DEGs,其中上调、下调表达的基因分别有22928、21735个;在相同时间点抗、感品种间的R_(1) vs S_(1)、R_(2) vs S_(2)、R_(3) vs S_(3)和R_(4) vs S_(4)这4个对比组中有1057个共有DEGs。GO富集分析结果表明:甘薯长喙壳菌侵染3 d时抗、感品种中富集到的前20的GO条目均主要为细胞成分,其次为生物过程和分子调控;隶属于生物过程的单一生物代谢过程、隶属于分子功能的氧化还原酶活性和四吡咯结合,以及隶属于细胞组分的质体和细胞外围等条目在抗、感品种中均被富集到。KEGG通路富集分析结果表明:在富集的前20的KEGG信号通路中,抗、感品种中的差异基因主要被富集到代谢途径、次级代谢产物的生物合成、碳代谢、苯丙烷生物合成、柠檬酸循环/三羧酸循环以及光合作用等通路上。此外,对长喙壳菌侵染甘薯块根的抗性相关基因进行了分析,共鉴定出221个抗性相关基因,其中激酶类(kinase,KIN)基因最多,有117个,占比为52.9%。In this study,the black rot disease resistant variety Nanjing 92 and the susceptible variety Xushu18 were used to screen the differentially expressed genes of sweetpotato in response to the infection of Ceratocystis fimbriata Ellis et Halsted by RNA-seq.The tubers of Nanjing 92 and Xushu18 which not infected by C.fimbriata were used as the control group,and the tubers infected by C.fimbriata for 1 d,3 d and 7 d were used as the treatment group for transcriptome sequencing respectively.The RNA-seq results showed that 36127 differentially expressed genes(DEGs)were found in disease resistant variety,including 17387 up-regulated genes and 18740 down-regulated genes.Meanwhile,44663 DEGs were found in susceptible variety,including 22928 up-regulated genes and 21735 down-regulated genes.Moreover,1057 common DEGs were found in the four comparison groups of R_(1) vs S_(1),R_(2) vs S_(2),R_(3) vs S_(3) and R_(4) vs S_(4).Go enrichment analysis of these DEGs showed that the Top 20 GO terms enriched in resistant and susceptible varieties mainly belonged to cellular components,followed by biological processes and molecular regulation.The GO term single biological metabolic process which belonged to biological processes,oxidoreductase activity and tetrapyrrole binding which belonged to molecular function,plastids and cell periphery which belonged to cellular component were enriched in both resistant and susceptible varieties.In addition,KEGG pathway enrichment analysis showed that the differential genes in resistant and susceptible varieties were mainly enriched in metabolic pathway,secondary metabolite biosynthesis,carbon metabolism,phenylpropionyl biosynthesis,citric acid cycle/tricarboxylic acid cycle,photosynthesis and so on.Meanwhile,221 resistance-related genes were identified,among which 117 were kinase(kin),accounting for 52.9%.
