摘  要:目的 探讨黑种草子提取物对慢性高原病大鼠心脏损伤的影响。方法 选择SD大鼠50只,随机分为平原组、高原组、硝苯地平组、黑种草子水提组和黑种草子醇提组,每组10只。高原组、硝苯地平组、黑种草子水提组和黑种草子醇提组模拟高原环境饲养30 d,构建慢性高原病模型。建模成功后,硝苯地平组予硝苯地平2.70 mg/kg灌胃,黑种草子水提组予黑种草子水提物150 mg/kg灌胃,黑种草子醇提组予黑种草子醇提物150 mg/kg灌胃,每天1次,连续15 d。高原组建模成功后继续在高原环境下饲养15 d,平原组在平原环境下饲养45 d,两组均不予药物干预。硝苯地平组、黑种草子醇提组和黑种草子水提组末次灌胃次日,高原组和平原组同期,检测各组肺动脉压和血清C反应蛋白(CRP)、血管紧张素原(AGT)、载脂蛋白C1(ApoC1)、胰岛素样生长因子1(IGF-1),并观察心脏病理形态变化。结果 与平原组比较,高原组肺动脉压和血清CRP、AGT、IGF-1水平显著升高,血清ApoC1水平显著降低(P均<0.05)。与高原组比较,硝苯地平组、黑种草子醇提组和黑种草子水提组肺动脉压显著降低(P均<0.05);硝苯地平组和黑种草子水提组血清ApoC1水平显著升高,血清CRP、AGT、IGF-1水平显著降低(P均<0.05);黑种草子醇提组血清IGF-1水平显著升高,血清CRP、AGT、ApoC1水平显著降低(P均<0.05)。与硝苯地平组比较,黑种草子醇提组和黑种草子水提组肺动脉压显著升高(P均<0.05);黑种草子醇提组血清CRP、ApoC1水平显著降低,血清AGT、IGF-1水平显著升高(P均<0.05);黑种草子水提组血清CRP水平显著降低,血清AGT、IGF-1水平显著升高(P均<0.05)。与黑种草子醇提组比较,黑种草子水提组肺动脉压显著升高(P<0.05),黑种草子水提组血清CRP、ApoC1水平显著升高,血清AGT、IGF-1水平显著降低(P均<0.05)。平原组心脏组织结构未见明显异常;高原组心脏组织结构紊乱,心肌纤维Objective To explore the effect of the extract of Nigellalandulifera Preynet Sint on heart injury in rats with chronic mountain sickness.Methods Fifty SD rats were randomly divided into five groups:plain group,plateau group,nifedipine group,water extract group,and alcohol extract group,with 10 rats in each group.The rats in the plateau group,nifedipine group,water extract group and alcohol extract group were fed in the simulated plateau environment for 30 days to construct the chronic mountain sickness models.After successful modeling,the rats in the nifedipine group were given nifedipine 2.70 mg/kg by gavage,the rats in the water extract group were given water extract of Nigellalandulifera Preynet Sint(150 mg/kg) by gavage,and the rats in the alcohol extract group were given alcohol extract of Nigellalandulifera Preynet Sint(150 mg/kg) by gavage,once a day,for 15 days.The rats in the plateau group continued to be raised in the plateau environment for 15 days,and the rats in the plain group were raised in the plain environment for 45 days.Rats in neither of the two groups received drug intervention.The pulmonary artery pressure and the content of serum C-reactive protein(CRP),angiotensinogen(AGT),apolipoprotein C1(ApoCl) and insulin-like growth factor-1(IGF-1) were measured in the plateau group,the plain group,the nifedipine group,the alcohol extract group,and the water extract group on the next day after the last gavage,and the pathological changes of heart were observed.Results Compared with the plain group,the plateau group had significantly higher pulmonary artery pressure(P<0.05),significantly higher serum CRP,AGT,and IGF-l levels(all P<0.05),and significantly lower serum ApoCl level(P<0.05).Compared with the plateau group,the nifedipine group,alcohol extract group and water extract group had significantly lower pulmonary artery pressure(all P<0.05),the nifedipine group and water extract group had significantly lower serum CRP,AGT,and IGF-1 levels(all P<0.05),and significantly higher serum ApoCl level(P<0.05
