摘　　要：目的探讨宫颈癌患者组织和血清中核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体相关蛋白的表达水平,分析NLRP3炎症小体在宫颈癌发生、进展和治疗中的潜在作用。方法选取2019年1月至2020年6月海南省人民医院收治的50例ⅠA~ⅡA期宫颈癌患者作为实验组,均行手术治疗,术中留取癌组织和未被肿瘤浸润的癌旁正常宫颈组织;同时选取50例健康女性作为对照组。所有研究对象均留取外周静脉血,采用酶联免疫吸附试验(ELISA)法检测血清中NLRP3、丙氨酸-丝氨酸半胱氨酸-1(ASC-1)、半胱氨酸酶-1(Caspase-1)、白介素-1β(IL-1β)、C-反应蛋白(CRP)、白介素-18(IL-18)和白介素-6(IL-6)的表达水平,采用旋转式血液流变仪检测红细胞沉降率(ESR),采用流式细胞仪检测白细胞数量;采用免疫组化染色法检测实验组癌组织和癌旁组织中NLRP3、ASC-1、Caspase-1的表达情况。结果实验组白细胞数量、ESR、CRP、IL-6、NLRP3、ASC-1、Caspase-1、IL-1β、IL-18水平明显高于对照组(P<0.05)。实验组癌组织中NLRP3、ASC-1和Caspase-1水平明显高于癌旁组织;NLRP3水平与CRP、IL-1β、IL-18、白细胞数量、ESR和IL-6呈显著正相关,IL-1β水平与白细胞数量、CRP、ESR、IL-18和IL-6呈显著正相关,IL-18的水平与白细胞数量、CRP、ESR、IL-1β和IL-6呈显著正相关(P<0.05)。结论NLRP3炎症小体的激活可能参与了宫颈癌的发生和进展,提示NLRP3炎症小体可作为治疗宫颈癌的潜在靶点。Objective To investigate the expression of nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3) inflammasome associated protein in tissues and serum of patients with cervical cancer,and to elucidate the potential role of NLRP3 inflammasome in the occurrence,progression and treatment of cervical cancer.Methods 50 patients with stage IA-IIA cervical cancer treated in Hainan General Hospital from January 2019 to June 2020 were selected as the experimental group,all were collected the tumor tissues and normal cervical tissues without tumor infiltration during the operation.50 healthy women were selected as the control group.Peripheral venous blood was collected from all subjects.The levels of NLRP3,alanine-serine-cysteine-1(ASC-1),Caspase-1,interleukin-1β(IL-1β),C-reactive protein(CRP),interleukin-18(IL-18) and interleukin-6(IL-6) in serum were detected by enzyme-linked immunosorbent assay(ELISA).Erythrocyte sedimentation rate(ESR) was measured by rotating blood rheometer and the number of white blood cells by flow cytometry.Immunohistochemical staining was used to detect the expression of NLRP3,ASC-1 and Caspase-1 in cancer tissues and adjacent tissues of the experimental group.Results The number of white blood cells,ESR,CRP,IL-6,NLRP3,ASC-1,Caspase-1,IL-1β and IL-18 levels in the experimental group were significantly higher than those in the control group(P<0.05).The levels of NLRP3,ASC-1 and Caspase-1 in the experimental group were significantly higher than those in the adjacent tissues.NLRP3 level was positively correlated with CRP,IL-1β,IL-18,white blood cell count,ESR and IL-6,and IL-1β level was positively correlated with white blood cell count,CRP,ESR,IL-18 and IL-6.The level of IL-18 was positively correlated with white blood cell count,CRP,ESR,IL-1β and IL-6(P<0.05).Conclusions The activation of NLRP3 inflammasome may be involved in the occurrence and progression of cervical cancer,suggesting that NLRP3 inflammasome can be used as a potential target for the treatment of cervical ca
