摘 要:通过简单的一步水热法制得二硫化钼纳米片(Molybdenum dioxide nanosheets,MoS_(2) NSs),利用X-射线衍射、X-射线光电子能谱、透射电子显微镜等对其结构及形貌进行表征。MoS_(2) NSs具有类过氧化物酶(Peroxidase-like,POD-like)活性,可催化过氧化氢(H_(2) O_(2))氧化无色的3,3′,5,5′-四甲基联苯胺(3,3′,5,5′-Tetramethylbenzidine,TMB)生成蓝色氧化产物(oxTMB),据此建立了测定H_(2) O_(2)的比色分析方法。优化了MoS_(2) NSs催化TMB-H_(2) O_(2)体系的显色反应条件,并进行了稳态动力学分析。结果表明,MoS_(2) NSs的酶催化反应遵循典型的Michaelis-Menten动力学模型。在最佳实验条件下,建立的比色体系检测H_(2) O_(2)的线性范围为10~50μmol/L,检出限(S/N=3)为1.0μmol/L。将本方法用于检测牛奶中的H_(2) O_(2),加标回收率为92.4%~106.9%。Molybdenum disulfide nanosheets(MoS_(2) NSs)were prepared by a simple one-step hydrothermal method,and their structure and morphology were characterized by X-ray diffraction(XRD),X-ray photoelectron spectroscopy(XPS)and transmission electron microscope(TEM).In the presence of hydrogen peroxide(H_(2)O_(2)),the obtained MoS_(2) NSs could catalyze the oxidation of colorless 3,3′,5,5′-tetramethylbenzidine(TMB)into blue oxidized products(oxTMB).On this basis,a colorimetric analysis method for determination of H_(2)O_(2) concentration was established.The effects of color reaction conditions of MoS_(2) NSs-TMB-H_(2)O_(2) on peroxidase-like activity of MoS_(2) NSs were investigated,and the steady-state kinetic analysis was carried out.The results showed that the enzymatic reaction of MoS_(2) NSs using TMB and H_(2)O_(2) as reaction substrates followed the typical Michaelis-Menten kinetic model.The linear range for colorimetric determination of H_(2)O_(2) was 10-50μmol/L,with a limit of detection(LOD,S/N=3)of 1.0μmol/L.The established method was used in detection of H_(2)O_(2) in milk samples and the recoveries of H_(2)O_(2) spiked in milk were 92.4%-106.9%,showing good practicability.
