摘　　要：目的探讨神经损伤诱导蛋白-1(NINJ1)在复发性流产(RSA)的表达变化以及对胎盘滋养层细胞的影响。方法收集2020年10月至2021年8月住院的复发性流产及计划外妊娠要求人工流产的正常胎盘绒毛组织各20例。分别检测其胎盘组织中NINJ1mRNA及NINJ1蛋白的表达;并采用转染法研究NINJ1对HTR-8/SVneo滋养层细胞增殖、迁移和侵袭的影响。结果RSA组患者胎盘绒毛组织中NINJ1表达显著上升。CCK8增殖实验结果表明,过表达NINJ1可显著降低细胞增殖率(P<0.05)。划痕实验结果表明,过表达NINJ1可显著降低细胞迁移率,而沉默NINJ1后细胞迁移率显著增加(P<0.05)。Transwell实验结果表明过表达NINJ1可显著降低侵袭细胞数,而沉默NINJ1后侵袭细胞数显著增加(P<0.05)。结论复发性流产患者胎盘绒毛组织中NINJ1mRNA及蛋白表达显著增加,NINJ1表达上调可显著抑制滋养层细胞增殖、迁移和侵袭能力,其参与复发性流产发生的具体调控机制值得深入研究。Objective To investigate the changes in expression of ninjurin-1 in recurrent spontaneous abortion and the effect on placental trophoblast cells.Methods Twenty cases each of recurrent spontaneous abortion and normal placental villi tissues hospitalized from October 2020 to August 2021 were collected.The expression of NINJ1 mRNA and NINJ1 protein in their placental tissues were detected by real-time PCR and Western blot,respectively,and the effect of NINJ1 on trophoblast cell proliferation,migration and invasion was investigated by transfection method.Results The expression of NINJ1 was significantly increased in placental villous tissues of RSA patients.the results of CCK8 proliferation assay showed that overexpression of NINJ1 significantly decreased the cell proliferation rate(P<0.05).The results of scratch assay showed that overexpression of NINJ1 significantly decreased the cell migration rate,while silencing of NINJ1 significantly increased the cell migration rate(P<0.05).Transwell assay showed that overexpression of NINJ1 significantly decreased the number of invasive cells,while silencing of NINJ1 significantly increased the number of invasive cells(P<0.05).Conclusion NINJ1 mRNA and protein expression were significantly increased in placental villous tissues of patients with recurrent spontaneous abortion,and upregulation of NINJ1 expression significantly inhibited trophoblast proliferation,migration and invasion ability.
