摘  要:目的探讨妊娠糖尿病(GDM)患者胎盘组织miR-103表达水平及其对滋养细胞自噬的影响。方法收集2019年1月至2019年12月40例GDM患者及40例正常孕妇胎盘组织。对HTR-8/SVneo细胞进行不同转染,分为Model组(高糖培养基培养),miR-NC组(转染miR-NC),miR-103组(转染miR-103 mimics),Si-miR-103组(转染miR-103 inhibitor)。流式细胞术检测细胞凋亡;透射电镜观察细胞超微结构;溶酶体红色荧光探针(lyso-tracker red)观察细胞自噬;反转录-聚合酶链反应(RT-PCR)检测胎盘组织、细胞miR-103表达;Western blot检测胎盘组织、细胞自噬相关蛋白3Ⅰ(LC3Ⅰ)、自噬相关蛋白3Ⅱ(LC3Ⅱ)、Beclin1、p62表达。结果与正常孕妇胎盘组织比较,GDM孕妇胎盘组织miR-103表达降低,LC3-Ⅱ/LC3-Ⅰ、Beclin1表达升高,p62表达降低(P<0.05)。与NC组比较,Model组miR-103表达降低,细胞凋亡率、自噬小体/细胞浆总面积比值升高,LC3-Ⅱ/LC3-Ⅰ、Beclin1表达上调,p62表达下调(P<0.05)。与miR-NC组比较,miR-103组细胞miR-103表达升高,细胞凋亡率、自噬小体/细胞浆总面积值降低,LC3-Ⅱ/LC3-Ⅰ、Beclin1表达下调,p62表达上调(P<0.05)。与miR-NC组比较,Si-miR-103组细胞miR-103表达下降,细胞凋亡率、自噬小体/细胞浆总面积值升高,LC3-Ⅱ/LC3-Ⅰ、Beclin1表达上调,p62表达下调(P<0.05)。结论miR-103在GDM胎盘组织中低表达,其高表达可抑制滋养细胞自噬。Objective To investigate the expression of miR-103 in placenta of gestational diabetes mellitus(GDM)and its effect on autophagy of trophoblast cells.Methods The placental tissues of 40 patients with GDM and 40 normal pregnant women from January 2019 to December 2019 were collected.HTR-8/SVneo cells were transfected and divided into model group(high glucose medium culture),miR-NC group(miR-NC transfection),miR-103 group(miR-103 mimics transfection)and si-miR-103 group(miR-103 inhibitor transfection).Apoptosis was detected by flow cytometry.The cell ultrastructure was observed by transmission electron microscope.Lysosomal red fluorescent probe was used to observe autophagy.The expression of miR-103 in placental tissues and cells was detected by reverse transcription polymerase chain reaction(RT-PCR).The expressions of autophagy related protein 3I(LC3I),autophagy related protein 3ii(LC3II),Beclin1 and p62 in placental tissue were detected by Western blot.Results Compared with the placental tissues of normal pregnant women,the expression of miR-103 decreased,the expression of LC3-II/LC3-I and Beclin1 increased,and the expression of p62 decreased in the placental tissues of GDM pregnant women(P<0.05).Compared with NC group,the expression of miR-103 decreased,the apoptosis rate and the ratio of autophagy body/total cytoplasmic area increased,the expression of LC3-II/LC3-I and Beclin1 increased,and the expression of p62 decreased in model group(P<0.05).Compared with miR-NC group,the expression of miR-103 in miR-103 group increased,the apoptosis rate and the ratio of autophagy body/total cytoplasmic area decreased,the expression of LC3-II/LC3-I and Beclin1 decreased,and the expression of p62 increased(P<0.05).Compared with miR-NC group,the expression of miR-103 decreased,the apoptosis rate and the ratio of autophagosome/total cytoplasmic area increased,the expression of LC3-II/LC3-I and Beclin1 increased,and the expression of p62 decreased in si-miR-103 group(P<0.05).Conclusion miR-103 is low expressed in GDM placenta,an
