摘 要:目的:研究胡桃醌对胃癌SGC-7901细胞增殖、侵袭和糖酵解的影响,并进一步探讨其分子机制。方法:MTT法和细胞克隆实验检测胡桃醌对SGC-7901细胞增殖的影响;流式细胞术检测胡桃醌对SGC-7901细胞周期的影响;划痕实验和transwell侵袭实验检测胡桃醌对SGC-7901细胞迁移和侵袭的影响;ELISA检测胡桃醌对SGC-7901细胞糖酵解的影响;Western blot检测糖酵解相关蛋白以及AKT/mTOR信号通路蛋白的变化。结果:胡桃醌能够有效抑制SGC-7901细胞的增殖,阻滞SGC-7901细胞于G_(0)/G_(1)期,能够显著抑制SGC-7901细胞的迁移和侵袭,降低SGC-7901细胞糖酵解反应,并且抑制AKT/mTOR信号通路的活性,与空白对照组相比均有显著性差异(P<0.05)。结论:胡桃醌能够抑制胃癌SGC-7901细胞增殖、侵袭和糖酵解,其机制与抑制AKT/mTOR信号通路有关。Objective:To investigate the effects of Juglone on proliferation,invasion and glycolysis of SGC-7901 cells,and to further explore its molecular mechanism.Methods:The effect of Juglone on the proliferation of SGC-7901 cells was determined by MTT assay and cell cloning assay;The effect of Juglone on the cell cycle of SGC-7901 was detected by flow cytometry;The effects of Juglone on the migration and invasion of SGC-7901 cells were examined by scratch assay and transwell invasion assay;The effect of Juglone on glycolysis of SGC-7901 cells was determined by ELISA;Western blot assay was used to detect the effects of glycolytic proteins and AKT/mTOR signaling proteins.Results:Juglone could inhibit the proliferation of SGC-7901 cells,induce the cell cycle arrest at G_(0)/G_(1) phase,significantly inhibit the migration and invasion of SGC-7901 cells,reduce the glycolytic reaction of SGC-7901 cells,and inhibit the activity of AKT/mTOR signaling pathway,which were significantly different from the blank control group(P<0.05).Conclusion:Juglone can inhibit the proliferation,invasion and glycolysis of SGC-7901 cells,and its mechanism is related to the inhibition of AKT/mTOR signaling pathway.
