基于GC-MS法分析二十五味珍珠丸中挥发油和脂溶性成分

摘　　要：目的:采用气相色谱-质谱(GC-MS)法分析5个厂家的7个批次二十五味珍珠丸中的挥发油和脂溶性成分,为后期有效成分研究奠定基础。方法:水蒸气蒸馏法提取二十五味珍珠丸粉末中的挥发油成分,溶剂萃取法提取二十五味珍珠丸粉末中的脂溶性成分,采用GC-MS法分离鉴定,利用NIST14谱库检索对各色谱峰结合有关图谱解析并加以确认,并采用峰面积归一法测定各个化合物的相对百分含量。结果:挥发油成分鉴定出111个化合物,共11类,为胺类,烷烃类、烯烃类、醇类、苯类、酚类、酮类、醛类、酯类和醚类,羧酸类,其中以醇类最多,其次为烯烃和酯类。脂溶性成分鉴定出168个化合物,共13类,为胺类,烯烃类、醇类、酯类、烷烃类、羧酸类、酚类、苯类、醛类、生物碱类、酮类、醚类,硫类,其中以醇类最多,其次为烯烃和酯类。结论:二十五味珍珠丸挥发油主要成分为丁香酚、肉豆蔻酸、甲苯等,脂溶性成分为亚油酸、肉豆蔻酸、棕榈酸等,可能来自于二十五味珍珠丸中的丁香、肉豆蔻、红花、西红花、余甘子等,有助于进一步明确二十五味珍珠丸的药效物质基础。Objective: To analyze the volatile oil and fat-soluble components in Twenty-five flavor Zhenzhu Pills of 7 batches from 5 manufacturers through Gas chromatography-mass spectrometry(GC-MS), which lays a foundation for the later study of effective components. Method: The volatile oil components in the powder of Twenty-five flavor Zhenzhu Pills were extracted by steam distillation, and the fat-soluble components were extracted by solvent extraction, which were isolated and identified by GC-MS. The chromatographic peaks were analyzed and confirmed by the combination of relevant spectrograms and NIST14library search, and the relative contents of each compound were detected by peak area normalization method. Result: 111 volatile oil components were identified, including 11 categories, such as amines, alkanes, alkanes, alcohols, benzene, phenols, ketones,aldehydes, esters, ethers and carboxylic acids, among which alcohols were the most, followed by olefins and esters. 168 fat-soluble components were identified, including 13 categories, such as amines, alkanes, alcohols, esters, alkanes, carboxylic acids, phenols,benzene, aldehydes, alkaloids, ketones, ethers and sulfur, among which alcohols were the most, followed by olefins and esters.Conclusion: TThe main volatile oil components of Twenty-five flavor Zhenzhu Pills are eugenol, myristic acid and toluene, and the fat-soluble components are linoleic acid, myristic acid and palmitic acid, which may come from Eugenia caryophyllata Thunb.,Myristica fragrans Houtt., Carthamus tinctorius L., Crocus sativus L. and Phyllanthus emblica L. in Twenty-five flavor Zhenzhu Pills. The results contribut to the further clarification of the pharmacodynamic material basis of Twenty-five flavor Zhenzhu Pills.
