摘 要:目的:研究健脑丸对东莨菪碱致学习记忆障碍模型小鼠的影响及其机制。方法:设空白、模型、阳性及健脑丸高、中、低剂量组,连续给药30天后,注射东莨菪碱诱导记忆障碍;Morris水迷宫测定小鼠空间学习记忆功能;HE染色观察脑组织病理变化,检测海马匀浆组织中Ach、AchE和ChAT含量。结果:与模型组比较,各组潜伏期值均明显小于模型组(P<0.05);健脑丸中剂量组目标象限路程百分比显著提高,ChAT、Ach水平显著提高,AchE显著降低(P<0.05)。与空白组比较,模型组神经元明显空泡化;与模型组比较,健脑丸能减少海马组织神经元空泡化。结论:健脑丸对东莨菪碱致学习记忆障碍模型小鼠空间学习及记忆能力具有改善作用,其机制可能与提高ChAT、Ach水平,降低AchE水平及保护海马神经元有关。Objective: To study the effect and mechanism of Jiannao Wan on scopolamine-induced learning and memory impairment in mice. Method: Control, model, positive groups, as well as high-, medium-and low-dose Jiannao Wan groups were set up. After 30 days of continuous administration, scopolamine was injected to induce memory impairment. The spatial learning and memory function of mice was measured by Morris water maze. The pathological changes of the brain tissue were observed by HE staining, and the contents of Ach, AchE and ChAT in hippocampal homogenate were detected. Result: Compared with the model group, the latency period of each group was significantly shorter(P < 0.05). In the medium-dose Jiannao Wan group, the percentage of target quadrant distance was increased significantly, with significant increased ChAT and Ach levels, and significant decreased ache level(P < 0.05). Compared with the control group, the neurons vacuolation was obvious in the model group.Compared with the model group, Jiannao Wan reduced the vacuolation of neurons in hippocampus. Conclusion: Jiannao Wan can improve the spatial learning and memory ability in the mouse model of scopolamine-induced learning and memory impairment.The mechanism may be related to increasing the levels of ChAT and Ach, reducing the level of AchE, and protecting hippocampal neurons.
