摘  要:SRO (similar to radical-induced cell death 1)是一类植物特异小分子蛋白家族,在植物生长发育和环境响应中发挥重要作用。然而SROs在大麦中仍缺乏系统鉴定。本研究从大麦中分离和鉴定了10个SRO家族成员(命名为HvSRO1~10)。系统发育和进化分析表明,SRO家族成员数目随着植物的分化和进化发生谱系特异性扩增,大麦SRO (HvSRO)可以分为3个亚组,与水稻OsSROs进化关系更近。基因结构显示,HvSRO基因不均等分布在6条染色体上,外显子-内含子数目差异较大,含有复杂的基因结构。多数大麦SRO家族成员特异靶向细胞核。从HvSRO基因的启动子中鉴定出许多激素、光和非生物胁迫应激响应顺式作用元件,它们与多种胁迫响应蛋白和mi RNA存在互作关系。RNA-seq分析表明SRO基因在大麦不同组织和发育阶段中广泛表达,且存在明显组织特异性特点,这些结果为进一步探究大麦SRO基因功能表达提供了理论依据,也为大麦抗性育种提供了潜在的基因资源。SRO(similar to radical-induced cell death 1) is a family of plant-specific small molecular proteins that plays an important role in plant growth and development and environmental response.However,SROs were still not systematically identified in barley.In this study,ten members of SRO family were isolated and identified from barley(named HvSRO1–10).Phylogenetic and evolutionary analyses showed that the number of SRO family members expanded with plant differentiation and evolution.HvSROs could be divided into three subgroups,whose evolutionary relationship was much closer with OsSROs.Gene structure showed that Hv SROs were unequally distributed on six chromosomes.Hv SROs had highly variable exon-intron numbers and contained a complex gene structure.Most HvSROs specifically targeted the nucleus.Many phytohormones,light and abiotic stress-responsive cis-acting elements were identified from the promoter of the Hv SROs,which interacted with a variety of stress-responsive proteins and miRNAs.RNA-seq analysis showed that Hv SROs were widely expressed in different tissues and developmental stages of barley and had distinct tissue-specific characteristics.These results provided a theoretical basis for further investigation of the expression pattern of the HvSROs and a potential genetic resource for barley resistance breeding.
