摘 要:目的观察和探讨摩腹手法对睡眠-觉醒相关神经递质及下丘脑Orexin系统的影响。方法将64只大鼠随机分为空白组、模型组、摩腹组和拮抗组各16只,采用自制改良平台水环境法对后三组大鼠进行原发性失眠造模,造模后空白组和模型组大鼠不作干预,摩腹组和拮抗组大鼠给予每日1次的摩腹治疗,其中拮抗组每日治疗前给予拮抗剂灌胃,干预持续14 d后,分析比较各组大鼠下丘脑、脊髓和结肠组织中Orexin-A及其m-RNA的表达水平,以及脑干、下丘脑组织中睡眠-觉醒相关神经递质的水平。结果与空白组相比,模型组大鼠脑干内DA、NE和ACH水平均有明显升高(P<0.05),脑干内5-HT水平明显降低(P<0.05);下丘脑和脊髓内Orexin-A水平均有明显升高(P<0.05),下丘脑、脊髓和结肠组织中prepro-Orexin-mRNA水平均有明显升高(P<0.05)。与模型组相比,摩腹组和拮抗组大鼠脑干内DA和NE水平均有明显降低(P<0.05),5-HT水平明显升高(P<0.05);下丘脑内Orexin-A和prepro-Orexin-mRNA水平均有明显降低(P<0.05)。结论摩腹手法可通过调节原发性失眠大鼠下丘脑内Orexin-A以及脑干内DA、NE和5-HT水平,起到调节大鼠睡眠和觉醒状态、治疗原发性失眠的作用。其中,下丘脑Orexin系统只是部分参与了摩腹手法对睡眠-觉醒相关神经递质的影响,且并不是唯一参与此过程的调节系统。Objective To observe and investigate the effects of abdominal rubbing on sleep-wake related neurotransmitters and hypothalamus Orexin system.Methods Sixty-four rats were randomly divided into blank group,model group,abdominal rubbing group and antagonistic group,with 16 rats in each group.A self-made improved platform water environment method was used to set up the primary insomnia latter model of three groups.After modeling,the rats in the blank group and model group were left without intervention.Rats in the antagonistic group and the abdominal rubbing group were given abdominal rubbing therapy once a day,and rats in the antagonistic group were given antagonist by gavage before daily treatment.After 14 days of intervention,the expression levels of Orexin-A and its mRNA in hypothalamus,spinal cord and colon,as well as the sleep-wake related neurotransmitters in brain stem and hypothalamus were analyzed and compared in each group.Results Compared with those of the blank group,the levels of dopamine(DA),norepinephrine(NE)and acetylcholine(Ach)in the brain stem of the model group were significantly increased(P<0.05),and the levels of 5-hydroxytryptamine(5-HT)in the brain stem were significantly decreased(P<0.05).The levels of Orexin-A in hypothalamus and spinal cord were significantly increased(P<0.05),and the levels of prepro-Orexin-mRNA in hypothalamus,spinal cord and colon were significantly increased(P<0.05).Compared with those of the model group,the levels of DA and NE in brain stem of rats in abdominal rubbing group and antagonistic group were significantly decreased(P<0.05),while the level of 5-HT was significantly increased(P<0.05).The levels of Orexin-A and prepro-Orexin-mRNA in hypothalamus were significantly decreased(P<0.05).Conclusion By regulating the levels of Orexin-A in the hypothalamus and DA,NE and 5-HT in the brain stem,abdominal rubbing manipulation can regulate the state of sleep and wake in rats and treat primary insomnia.Among them,the Orexin system of the hypothalamus is partially involved
