摘 要:目的制备麸炒枳壳标准汤剂,进行质量及抗氧化活性研究。方法制备15批麸炒枳壳标准汤剂,采用高效液相色谱法测定标准汤剂中柚皮苷和新橙皮苷的含量,建立标准汤剂的指纹图谱,对共有峰相对峰面积进行聚类分析和主成分分析。测定枳壳标准汤剂的总抗氧化能力(FRAP法)、DPPH自由基清除能力及ABTS自由基清除能力。结果15批麸炒枳壳标准汤剂的出膏率为10.42%~16.51%;柚皮苷含量为7.27%~12.81%,转移率为19.04%~42.26%;新橙皮苷含量为4.98%~8.72%,转移率为13.87%~24.19%。指纹图谱确定了12个共有峰,相似度均大于0.95,聚类分析将15批麸炒枳壳分为两类。主成分分析得到3个主成分,累积贡献率为83.831%。枳壳标准汤剂总抗氧化能力值为[(0.1847±0.0258),100μg·mL^(-1)],ABTS自由基清除能力可达[(80.42±11.93)%,2500μg·mL^(-1)],DPPH自由基清除能力可达[(94.98±8.65)%,2500μg·mL^(-1)]。结论麸炒枳壳标准汤剂制备规范,含量和指纹图谱测定方法精密度、稳定性和重复性良好,可为麸炒枳壳配方颗粒质量控制提供参考,麸炒枳壳标准汤剂有良好的体外抗氧化效果。Objective To prepare Aurantii Fructus Praeparatus standard decoction,and determine its quality and antioxidant activity.Methods The content of naringin and nehesperidin in the standard decoction was determined by HPLC.The fingerprint of the standard decoction was established.Cluster analysis and principal component analysis were performed on the relative peak area of common peaks.Total antioxidant capacity(FRAP),DPPH free radical scavenging capacity and ABTS free radical scavenging capacity of Aurantii Fructus Praeparatus standard decoction were determined.Results The extraction rate of 15 batches of Aurantii Fructus Praeparatus standard decoction was 10.42%~16.51%.The content of naringin was 7.27%~12.81%,with the transfer rate of 19.04%~42.26%.The content of new hesperidin was 4.98%~8.72%,with the transfer rate of 13.87%~24.19%.The fingerprint determined 12 common peaks,similarity being greater than 0.95.The 15 batches of Aurantii Fructus Praeparatus were divided into 2 categories by cluster analysis.Three principal components were obtained from principal component analysis,with a cumulative contribution rate of 83.831%.The total antioxidant capacity(Trolox Nmol)of Aurantii Fructus Praeparatus standard decoction was[(0.1847±0.0258),100μg·mL^(-1)],and ABTS radical scavenging ability was[(80.42±11.93)%,2500μg·mL^(-1)].DPPH radical scavenging activity was[(94.98±8.65)%,2500μg·mL^(-1)].Conclusion Aurantii Fructus Praeparatus standard decoction is prepared,and the determination is precise,stable and repeatable,which provides reference for the quality control of Aurantii Fructus Praeparatus formula particles.Aurantii Fructus Praeparatus standard decoction shows good antioxidant effect in vitro.
