摘　　要：目的建立高效液相色谱法同时测定华北獐牙菜中獐牙菜苦苷、龙胆苦苷、当药苷、芒果苷、异荭草素的含量。方法采用Waters SunFire C_(18)柱(4.6 mm×250 mm,5μm),甲醇-0.05%磷酸溶液为流动相进行梯度洗脱,流速1.0 mL·min^(-1),检测波长254 nm,进样量10μL,柱温30℃。结果獐牙菜苦苷、龙胆苦苷、当药苷、芒果苷、异荭草素进样量分别在0.0546~1.0930μg(r=0.9993)、0.1910~3.8200μg(r=0.9996)、0.1089~2.1780μg(r=0.9995)、0.0584~1.1690μg(r=0.9999)、0.0528~1.0570μg(r=0.9992)与峰面积呈良好的线性关系,加样回收率分别为97.62%、97.04%、97.68%、95.66%、96.75%,RSD分别为1.3%、0.79%、0.53%、0.79%、0.92%(n=6);3批样品中龙胆苦苷含量最高。结论该方法简便可行,精密度高,重复性好,可用于华北獐牙菜的质量控制。Objective To establish an HPLC method for the simultaneous determination of swertiamarin,gentiopicroside,sweroside,mangiferin and isoorientin in Swertia wolfangiana Grun.Methods Waters SunFire C_(18) column(4.6 mm×250 mm,5μm)was used with methanol-0.05% phosphoric acid solution as the mobile phase at 1.0 mL·min^(-1).The detection wavelength was 254 nm,the sample volume was 10μL,and the column temperature was 30℃.Results The linearity for swertiamarin,gentiopicroside,sweroside,mangiferin and isoorientin were 0.0546~1.0930μg(r=0.9993),0.1910~3.8200μg(r=0.9996),0.1089~2.1780μg(r=0.9995),0.0584~1.1690μg(r=0.9999),and 0.0528~1.0570μg(r=0.9992).The recoveries were 97.62%,97.04%,97.68%,95.66%,and 96.75%.RSDs were 1.3%,0.79%,0.53%,0.79%,and 0.92%(n=6).Gentiopicroside had the highest content among the 3 samples batches.Conclusion The method is simple,feasible,accurate and reproducible,which can be used for the quality control of Swertia wolfangiana Grun..
