摘  要:目的:探究绿萝花黄酮对糖尿病周围神经病变(DPN)小鼠运动神经传导速度以及NF-κB信号通路的影响。方法:将90只C57BL/6小鼠随机分为空白组10只和高脂高糖组80只,采用高脂高糖饲料联合链脲佐菌素(STZ)建立DPN模型,造模成功的小鼠随机分为模型组、阿卡波糖组(150 mg/kg)和绿萝花总黄酮高、中、低剂量组(200、100、50 mg/kg)各13只,治疗不同时间(10、20、40 d)时比较各组小鼠体重、血糖、坐骨神经运动神经传导速度(MNCV)和感觉神经传导速度(SNCV)及热痛反应敏感程度,血清TNF-α、IL-1β水平,坐骨神经组织形态学和NF-κB信号通路体内表达水平;培养乳鼠坐骨神经原代施万细胞,分为正常组、LPS组、LPS+阿卡波糖组(40μg/ml)、LPS+绿萝花总黄酮高、中、低剂量组(80、40、20μg/ml),药物干预后检测不同时间(12、24、48 h)细胞上清液中TNF-α、IL-6水平和NF-κB信号通路蛋白、基因表达水平。结果:体内实验结果表明,治疗40 d,与模型组比较,阿卡波糖组、绿萝花黄酮高、中剂量组小鼠体重逐渐升高(P<0.05),血糖水平逐渐降低(P<0.05),热痛反应时间、坐骨神经MNCV、SNCV逐渐缩短(P<0.05),坐骨神经病理改变得到不同程度改善;体内外实验结果均显示,阿卡波糖以及高、中剂量绿萝花黄酮干预后TNF-α、IL-1β水平逐渐降低(P<0.05),NF-κB p65蛋白表达量和p-IKKβ/IKKβ、p-IKKα/IKKα比值以及NF-κB、IKKβ、IKKα、NF-κB p65的mRNA相对表达量均逐渐减少(P<0.05)。结论:绿萝花黄酮可降低DPN小鼠血糖水平,改善运动、感觉神经传导速度,减轻坐骨神经炎症反应,可能与抑制NF-κB信号通路相关。Objective:To explore the effects of Flavonoids from Scindapsus Aureus on motor nerve conduction velocity and NF-κB signaling pathway in mice with diabetic peripheral neuropathy(DPN).Methods:90 C57BL/6 mice were randomly divided into blank group(10 mice)and high-fat and high-glucose group(80 mice).DPN model was established with high-fat and high-glucose diet combined with streptozotocin(STZ).Mice with successful modeling were randomly classified into model group,acarbose group(150 mg/kg),and high-dose,middle-dose and low-dose total flavonoids from Scindapsus Aureus groups(200,100,50 mg/kg),with 13 mice in each group.The body mass,blood glucose,motor nerve conduction velocity(MNCV)and sensory nerve conduction velocity(SNCV)of sciatic nerve,sensitivity of thermal pain response,levels of serum TNF-αand IL-1β,sciatic nerve histomorphology and expression level of NF-κB signaling pathway in vivo were compared among the groups at different treatment time points(10,20,40 d).Primary Schwann cells of sciatic nerve of suckling mice were cultured and divided into normal group,LPS group,LPS+acarbose group(40μg/ml)and LPS+high-dose,middle-dose and low-dose total Flavonoids from Scindapsus Aureus groups(80,40,20μg/ml).The levels of TNF-αand IL-6 and expression levels of NF-κB signaling pathway protein and gene in cell supernatants at different time points(12,24,48 h)were detected after drug intervention.Results:In vivo experiments showed that during 40 d of treatment,the body mass of the mice in the acarbose group and the high-dose and middle-dose total flavonoids groups was increased gradually compared with that in the model group(P<0.05),and the blood glucose level was gradually decreased(P<0.05),and the thermal pain response time,MNCV and SNCV were gradually shortened or reduced(P<0.05),and the pathological changes of sciatic nerve were improved to varying degrees.In vitro and in vivo experiments showed that after intervention with acarbose and high-and middle-dose total flavonoids from Scindapsus Aureus,the levels of
