摘  要:目的:运用网络药理学方法探讨黄柏-土茯苓治疗慢性前列腺炎的作用机制。方法:根据药物性质和口服生物利用度,利用中药系统药理学数据库与分析平台(TCMSP)收集两味药的化合物和有效靶点进行分析,采用Cytoscape 3.9.0构建化合物-靶点网络结构;从GeneCards中获得慢性前列腺炎相关基因的靶点,结合黄柏-土茯苓有效成分对上述靶点进行韦恩分析,并筛选出交集靶点。利用String数据库建立蛋白质-蛋白质相互作用网络;在DAVID数据库中进行基因本体(GO)功能富集分析和与京都基因和基因组数据库(KEGG)通路富集分析。结果:获得活性化合物36个,药物靶点218个,疾病靶点2204个,交集靶点157个。获得白细胞介素(Interleukin,IL)-4、IL-6、IL-10、C-C基序趋化因子配体2(C-C Motif Chemokine Ligand 2,CCL2)、C-X-C基序趋化因子配体(C-X-C Motif Chemokine Ligand,CXCL)2、CXCL10、肿瘤坏死因子(Tumor Necrosis Factor,TNF)等48个关键靶点,KEGG通路富集分析结果显示主要涉及TNF信号通路、白细胞介素-17信号通路和核转录因子-κB(Nuclear Transcription Factor-κB,NF-κB)信号通路。结论:初步分析了黄柏-土茯苓的药理网络,揭示了两味中药治疗慢性前列腺炎的潜在靶点及相关信号通路,为后续深入研究提供了一定的参考。Objective:To explore the mechanism of Huangbo(cortex phellodendri chinsis)-Tufuling(smilacis glabrae rhizoma)in treating chronic prostatitis by network pharmacology.Methods:According to the drug properties and oral bioavailability,the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)was used to collect the compounds and effective targets of the two herbs for analysis,Cytoscape 3.9.0 was used to construct the network structure of compound-target;the targets of CP-related genes were obtained from the Human Gene Database(GeneCards).The above targets were subjected to Venny analysis with the active ingredients of Huangbo-Tufuling,and the intersection targets were identified.The target protein-protein interaction network was established by the String database.GO analysis of gene function and KEGG pathway enrichment analysis were performed in the DAVID database.Results:The 36 active compounds were obtained,with 218 drug targets,2204 disease targets,and 157 intersection targets.Forty-eight key targets were obtained,including IL-4,IL-6,IL-10,CCL2,CXCL2,CXCL10,and TNF.The results of KEGG pathway enrichment analysis showed that it mainly involved TNF signaling pathway,IL-17 signaling pathway,and NF-kappa B signaling pathway.Conclusion:The pharmacological network of Huangbo-Tufuling was preliminarily analyzed,and the potential targets and signaling pathways of the two medicines in the treatment of chronic prostatitis were revealed,providing some reference for subsequent in-depth study.
