摘  要:红葱(Eleutherine bulbosa)在化学成分和药理活性开发方面具有重要价值,但是分子生物学研究较少。本研究以红葱花期全株为研究对象,利用PacBio Sequel^(TM)测序平台进行红葱根、鳞茎、叶片以及花等组织部位混合样品的全长转录组测序。测序数据经过校正后,得到高质量Isoforms序列182873条,利用Nr、Swiss Prot、KOG和KEGG四个数据库进行比对获得106318条全长转录本,分别有105734(Nr)、90207(Swissport)、77965(KOG)和52047(KEGG)条转录本获得功能注释。其中KEGG数据库分析结果共获得了132条代谢途径,主要有次生代谢产物合成、抗菌素合成、糖和淀粉代谢等。高级注释共获得105976个CDS,共预测到3257个转录因子。结构分析共鉴定到39666个SSRs,对没有注释到四大数据库的全长转录序列进行lncRNA分析,共预测到6394个lncRNA。本研究为红葱二代转录组测序数据拼接提供参考模板,为红葱全长基因克隆、基因功能分析和活性成分合成机制解析等提供理论基础。Eleutherine bulbosa is an important medicinal plant in chemical composition development and pharmacological activity study areas,but its research foundation of molecular biology is relatively weak.The full-length transcriptome sequencing and functional annotation analysis of root,bulb,leaf and flower of Eleutherine bulbosa were performed using Pac Bio Sequel^(TM)sequencing platform.According to the full-length transcriptome data,182873 high-quality isoforms were obtained in total.Moreover,the unigenes were further annotated into the public databases of Nr,SwissProt,KOG and KEGG.The results showed that Nr annotation revealed 105734Unigenes,Swissport annotation revealed 90207 Unigenes and KOG annotation revealed 77965 Unigenes.Among them,KEGG analysis found that 52047 Unigenes were annotated into 132 metabolic pathways,including biosynthesis of secondary metabolites,biosynthesis of antibiotics,starch and sucrose metabolism and so on.A total of 105976 CDS and 3257 transcription factors were predicted by the advanced annotation.In addition,39666SSRs were identified by structural analysis,and 6394 lncRNAs were predicted.This study provides a reference template for the splicing of transcriptome sequencing data of the second generation of shallot,and provides a theoretical basis for full-length gene cloning,gene function analysis and synthesis mechanism analysis of active components of Eleutherine bulbosa.
