摘 要:本研究以‘北农青贮368’、‘京科青贮516’、‘京科968’、‘东科301’为研究对象,采用单因素和响应面试验优化青贮玉米中的可溶性糖的提取工艺,通过酶-DNS(3,5-Dinitrosalicylic acid)法与近红外扫描法测定青贮玉米淀粉含量,旨在获得青贮玉米中淀粉含量测定的最佳方法。结果表明:(1)可溶性糖提取的最佳工艺为料液比1∶58(g/mL),提取温度为86℃,提取时间为61 min,此条件下可溶性糖含量为11.25%。(2)酶-DNS法测定青贮玉米品种及其全株/秸秆淀粉含量分别为‘东科301’(43.95%/6.72%)、‘北农青贮368’(29.65%/6.28%)、‘京科青贮516’(31.09%/4.58%)、‘京科968’(39.22%/5.87%),相对标准偏差为0.04%~0.45%。(3)近红外扫描法青贮玉米品种及其全株/秸秆淀粉含量分别为‘东科301’(42.96%/5.88%)‘、北农青贮368’(28.39%/6.06%)、‘京科青贮516’(30.22%/4.91%)、‘京科968’(36.15%/5.94%)。两种方法相比差异不显著(P>0.05),酶-DNS法与近红外扫描法均可用于青贮玉米中淀粉含量的测定。研究结果可为快速、便捷测定青贮玉米中淀粉含量提供参照依据。In this study we took’Beinong Silage 368’,’Jingke Silage 516’,’Jingke 968’,and’Dongke 301’as research objects.The extraction process of water soluble carbohydrate(WSC)in silage corn was optimized by single factor and response surface experiments,the enzyme-DNS method and near infrared reflectance spectroscopy method were used to determine starch content of silage corn to study a better method.The results showed that:(1)The best process for water soluble carbohydrate extraction was that the material-liquid ratio was 1∶58(g/m L),the extraction temperature was 86℃,the extraction time was 61 minutes,and under these conditions,the water soluble carbohydrate content was 11.25%.(2)Enzyme-DNS method to determine the starch content of silage corn whole plant/straw was’Dongke 301’(43.95%/6.72%),’Beinong Silage 368’(29.65%/6.28%),’Jingke Silage 516’(31.09%/4.58%),’Jingke 968’(39.22%/5.87%),and the relative standard deviation between repeats was 0.04%~0.45%.(3)The starch content of the whole plant/straw of corn silage scanned by near infrared reflectance spectroscopy was’Dongke 301’(42.96%/5.88%),’Beinong Silage 368’(28.39%/6.06%),’Jingke Silage 516’(30.22%/4.91%),’Jingke 968’(36.15%/5.94%).There was not significant difference between the two methods(P>0.05),and the enzyme-DNS method and near infrared reflectance spectroscopy method could be used for the determination of starch content in silage corn.The results can provide reference for rapid and convenient determination of starch content in silage corn.
