摘  要:为寻找提高青冈属植物扦插生根率的有效途径,本研究探索了单宁对扦插生根效果的影响。以3种青冈属植物枝条为试验材料,用福林酚法对枝条中的单宁含量进行测定,并用PVP、PVPP和丙酮对单宁进行了脱除,进而应用到扦插试验中。结果表明,3种青冈属植物枝条中单宁含量最高的为赤皮青冈,达到了90.55 mg/g。对提取液中的单宁进行脱除处理后发现,PVP和PVPP对单宁的脱除效果较好,不同树种的脱除率均在40%以上和60%以上;对枝条中的单宁进行脱除处理后发现,用10%丙酮溶液浸泡枝条24 h后单宁脱除效果最好,PVPP次之,而PVP脱除效果最差。利用低浓度的PVPP和丙酮溶液对枝条中的单宁进行脱除后再进行扦插,结果发现30%丙酮溶液浸泡2 h后扦插愈伤率和生根率明显高于其他处理,3个树种最高生根率分别达到了41.11%、34.44%和25.56%,说明用30%丙酮浸泡插穗2 h对提高扦插生根率有促进作用。研究结果为青冈属植物扦插繁育技术的提升提供了参考。In order to find an effective way to improve the rooting rate of Cyclobalanopsis after cuttings,this study explored the effect of tannin on the rooting effect of cuttings.In this study,the branches of three species of Cyclobalanopsis were used as test materials.First,the tannin content in the branches was measured by the Folin-phenol method,and then the tannins were removed by PVP,PVPP and acetone.The treated shoots were finally used in cutting experiments.The results showed that,among the three species of Cyclobalanopsis,the branch with the highest tannin content was Cyclobalanopsis glauca,reaching 90.55 mg/g.The experimental results of removing tannins in the extract showed that PVP and PVPP had better removal effects on tannins,and the removal rates of different species reached 40%and 60%,respectively.The experimental results of removing tannins from the branches showed that the removal effect of tannins was the best when the branches were soaked in 10%acetone solution for24 hours,followed by PVPP,and the removal effect of PVP was the worst.When the tannins in the branches were removed with low concentration of PVPP and acetone solution before cutting,the results showed that the callus rate and rooting rate of the cuttings after soaking in 30%acetone solution for 2 h were significantly higher than other treatments.The highest rooting rates of the three tree species reached 41.11%,34.44%and 25.56%,respectively,indicating that soaking cuttings with 30%acetone for 2 h could promote the rooting rate of cuttings.The results of this study provide a reference for the improvement of cutting propagation technology of Cyclobalanopsis.
