摘  要:细胞质雄性不育(cytoplasmic male sterility,CMS)被广泛应用于作物杂交种的生产。CMS通常由线粒体基因组中新的开放阅读框(open reading frame,ORF)所控制。本研究对细胞质雄性不育系种质HNUCA00HG的线粒体基因组进行了测序和分析,得到其基因组大小为564124 bp,比已经发表的不育系138A(504210 bp)和FS4401(507452 bp)分别长59915和56673 bp。共注释出77个已知功能基因,包括39个编码蛋白基因,33个编码tRNA基因和5个编码核糖体RNA基因。比较基因组和系统发育分析表明,这三个不育系的线粒体可能来自同一个祖先,但相互间存在明显差异。与HNUCA00HG线粒体相比,FS4401线粒体缺少两个已知的功能基因rps7和rps14,而138A不育系缺少rps7、rps14、atp4、atp8和mttB。初步预测了五个ORFs(ORF133a,ORF110a,ORF107a,ORF109a和ORF111a)为HNUCA00HG潜在的不育候选基因。因此,进一步解析辣椒细胞质雄性不育和杂交种培育提供了重要资源。Cytoplasmic male sterility(CMS)is widely used in the production of crop hybrids.CMS is usually controlled by mitochondrial genomic novel open reading frame(ORF).In this study,the mitochondrial genome of the cytoplasmic male sterile line germplasm HNUCA00HG was sequenced and analyzed,obtaining a genome size of 564124 bp,which is 59915 bp and 56673 bp longer than the published sterile lines 138A(504210 bp)and FS4401(507452 bp),respectively.A total of 77 known functional genes were annotated,including 39 genes encoding proteins,33 genes encoding tRNAs and 5 genes encoding ribosomal RNAs.Comparative genomic and phylogenetic analyses indicated that the mitochondria of these three sterile lines may be from the same ancestor,but are distinctly different from each other.Compared to the mitochondria of HNUCA00HG,FS4401 mitochondria lacked two known functional genes,rps7 and rps14,while 138A sterile lines lacked rps7,rps14,atp4,atp8 and mttB.Five ORFs(ORF133a,ORF110a,ORF107a,ORF109a and ORF111a)as potential sterility candidate genes for HNUCA00HG.The results of these studies provide important resources for further resolution of cytoplasmic male sterility and hybrid breeding in pepper.
