摘  要:为解析NtHD20(LOC107765920)基因对烟草腺毛发育的功能,将NtHD20在烟草(Nicotiana tabacum cv.Xanthin)中过表达,观察腺毛形态及数量变化,检测腺毛密度调控基因表达水平,测定内源激素含量。结果表明,NtHD20在烟草花中表达量最高,根中最低,表达具有组织特异性;过表达植株中烟草腺毛密度显著降低,腺毛密度正调控基因NtMYB123-like(LOC107773379)、NtWo-like(LOC107766155)与NtabSPL6-1(LOC1-07789814)表达量分别为野生型植株的0.14、0.48与0.49倍,负调控基因NtCycB2(Nitab4.5-0014697g0010.1)为野生型植株1.77倍,同时烟草ABA与JA含量显著降低,分别为野生型植株的0.56与0.16倍。表明NtHD20是烟草腺毛发育的负调控因子,可能通过ABA与JA两种激素协同介导下调腺毛发育重要基因(abSPL6-1,MYB123,Wo),同时上调细胞周期蛋白基因NtCycB2,从而降低烟草腺毛密度。To analyze the function of NtHD20(LOC107765920)gene on the development of tobacco glandular trichomes,NtHD20 was overexpressed in tobacco(Nicotiana tabacum cv.Xanthin),the morphology and quantity of glandular trichomes were observed,the expression levels of genes regulated by glandular trichomes density were detected,and the content of endogenous hormones was determined.The results showed that the expression of NtHD20 was the highest in tobacco flowers and the lowest in roots,and the expression was tissue-specific;the density of tobacco glandular trichomes was significantly reduced in overexpression plants,and the glandular trichomes density positively regulated genes NtMYB123-like(LOC107773379),NtWo-like(LOC107766155)and NtabSPL6-1(LOC107789814).The expression levels were 0.14,0.48 and 0.49 times that of wild-type plants,respectively,and the negative regulatory gene NtCycB2(Nitab4.5-0014697g0010.1)was 1.77 times that of wildtype plants.At the same time,the contents of ABA and JA in tobacco were significantly reduced,which were 0.56and 0.16 times that of wild-type plants,respectively.Suggest that NtHD20 is an important negative regulator of glandular trichomes development in tobacco.It may be mediated by two hormones,ABA and JA,to down-regulate important genes for glandular trichomes development(abSPL6-1,MYB123,Wo)and up-regulate the cyclin gene NtCycB2,thereby reducing tobacco glandular trichomes density.
