摘  要:沙棘是一种药食同源的植物,营养物质丰富,包括维生素类、黄酮类、挥发油类、甾醇类、多糖以及微量元素等,其中沙棘黄酮可以通过抗氧化,清除机体内多余活性氧。为了探讨沙棘黄酮联合规律运动通过FOXO信号通路改善AD模型大鼠学习与记忆能力。本研究选取60只健康雄性SD大鼠,体重在(220±10)g,设置5个组别,除了正常对照组(Con组)外,其他各组大鼠均进行D-半乳糖衰老造模。正常对照组和模型组(M组)大鼠正常饲养,规律运动组(G组)大鼠进行有氧跑台训练,沙棘黄酮组(S组)大鼠灌胃沙棘黄酮组,沙棘黄酮联合规律运动(GS组)大鼠同时进行有氧跑台训练和灌胃沙棘黄酮组治疗。有氧运动跑台训练每天训练15 min,每周5次;沙棘黄酮每天灌胃4.0 g/kg,每天灌服1次,整个训练周期持续12周。研究结果表明,沙棘黄酮联合规律运动能显著改善大鼠学习记忆能力;Real-Time PCR和Western blotting检测发现,沙棘黄酮联合规律运动能显著影响Bcl-2、Bax、Caspase-3以及FOXO-3α mRNA和蛋白水平表达情况,且与模型组相比,差异显著(P<0.05)。本研究结果初步表明,沙棘黄酮联合规律运动通过FOXO信号通路改善AD大鼠记忆与学习能力。Sea buckthorn(Hippophae rhamnoides)is a plant with medicinal and edible homology.It is rich in nutrients,including vitamins,flavonoids,volatile oils,sterols,polysaccharides and trace elements.Sea buckthorn flavonoids can remove excess reactive oxygen species in the body through oxidation resistance.In order to explore the sea buckthorn flavonoids joint regular exercise improves the learning and memory ability of AD model rats through FOXO signaling pathway.In this study,60 healthy male SD rats weighing between(220±10)g were selected,and five groups were set up.Except for the normal control group(Group C),the aging models of D-galactose were made in other groups.Rats in the normal control group and model group(Group M)were fed normally,rats in the regular exercise group(Group G)were given aerobic treadmill training,rats in the sea buckthorn flavone group(Group S)were given intragastric administration of sea buckthorn flavone group,and rats in the sea buckthorn flavone combined with regular exercise(Group GS)were given aerobic treadmill training and intragastric administration of sea buckthorn flavone group at the same time.Aerobic exercise treadmill training 15 minutes a day,5 times a week;Sea buckthorn flavone was administered by gavage of 4.0 g/kg per day,once a day,and the whole training cycle lasted for 12 weeks.The results showed that sea buckthorn flavonoids combined with regular exercise could significantly improve the learning and memory ability of rats;Real time PCR and Western blot showed that the combined regular exercise of sea buckthorn flavonoids could significantly affect Bcl-2,Bax,Caspase-3 and FOX O-3α.The expression of mRNA and protein levels was significantly different from that of the model group(P<0.05).The results of this study preliminarily show that sea buckthorn flavonoids combined with regular exercise can improve the memory and learning ability of AD rats through FOXO signaling pathway.
