表达小黄鱼生长激素的转基因家蚕品系构建

摘  要:构建转小黄鱼生长激素基因的家蚕新品系,外源表达鱼生长激素。通过检索小黄鱼转录组序列,获得小黄鱼生长激素和转铁蛋白的编码序列,设计并合成以接头序列连接,N端为生长激素、C端为转铁蛋白的融合蛋白编码框。利用piggyBac转座子介导的转基因方法,将融合基因转入家蚕基因组中,转育得到可表达小黄鱼生长激素且人工饲料摄食性较好的转基因家蚕品系皓月-Fc,蛹体中小黄鱼生长激素表达量最高约为0.27 ng/mg,蚁蚕24 h疏毛率约为85%。构建的转基因家蚕品系为下一步小黄鱼功能性饲料添加剂的研发打下基础。A transgenic silkworm variety inserted with the growth hormone(GH) of little yellow croaker(Larimichthys polyactis) was constructed to exogenously express the fish GH in silkworm bioreactor. The coding sequences of fish GH and transferrin were obtained by searching the transcriptome data of little yellow croaker. A hybrid gene encoding fusion protein was designed and synthesized, consisting of N terminal GH, C terminal transferrin and a linker sequence. The hybrid gene was introduced into silkworm genome with the help of piggyBac transposon. A new transgenic silkworm variety Haoyue-Fc, which can express GH of little yellow croaker and has good artificial diet intake, was obtained. The maximum expression of the fish GH in the pupa was about 0.27 ng/mg, and the setae dispersion rate in 24 h was about 85%. The successful breeding of the transgenic silkworm variety set a good foundation for further development of fish functional diet additives.
