白僵蚕三萜诱导人胃癌细胞SGC-7901 G_(1)期阻滞及凋亡的研究

摘　　要：采用MTT和流式细胞术等方法,研究白僵蚕三萜对人胃癌细胞SGC-7901生长的体外抑制作用及其作用机制。结果显示,白僵蚕三萜体外能有效抑制SGC-7901、人非小细胞肺癌细胞A549、人结肠癌细胞HCT116和人宫颈癌细胞HeLa增殖,其中对SGC-7901细胞生长的抑制作用最强,IC_(50)为77.08μg/mL,但对正常细胞(人胚肾细胞HEK293)生长无明显抑制作用;白僵蚕三萜能诱导SGC-7901细胞形态异常、贴壁细胞数量减少、在细胞G_(1)期阻滞,且诱导作用均呈浓度依赖性;75.0、87.5、100.0μg/mL白僵蚕三萜处理SGC-7901细胞48 h,均能引起细胞总凋亡率、早期凋亡率和晚期凋亡率显著增加,细胞内活性氧水平显著提高,线粒体膜电位显著降低。结果表明,白僵蚕三萜通过促进细胞凋亡、诱导细胞G_(1)期阻滞、促进细胞线粒体膜电位去极化,以及诱导细胞活性氧产生,发挥体外抑制SGC-7901细胞生长的作用。In vitro inhibitory effect of triterpenoids of Bombyx batryticatus on human gastric cancer cell SGC-7901 was determined through MTT assay and flow cytometry methods,and its possible mechanism was discussed in this paper.The results showed that triterpenoids of Bombyx batryticatus could inhibit the proliferation of SGC-7901,human non-small cell lung cancer cell A549,human colon cancer cell HCT116 and human cervical carcinoma cancer cell HeLa in vitro,and had nearly no inhibitory effect on normal human embryonic kidney cell HEK293.Among them,the inhibitory effect to SGC-7901 cell was the strongest,with an IC_(50) of 77.08μg/mL.Besides,triterpenoids of Bombyx batryticatus induced abnormal morphology of SGC-7901 cell,decreased the number of adherent cells,induced G_(1) arrest of SGC-7901 cell,and exhibited a concentration-dependent inhibitory effect.Moreover,after the treatment with Bombyx batryticatus triterpenoids of 75.0,87.5 and 100.0μg/mL for 48 h,total apoptosis rate,early apoptosis rate and late apoptosis rate were significantly increased,the intracellular reactive oxygen species level was significantly increased,and the mitochondrial membrane potential was significantly decreased.In conclusion,triterpenoids of Bombyx batryticatus inhibited the proliferation of SGC-7901 cell in vitro by promoting cell apoptosis,inducing cell G_(1) arrest,promoting depolarization of cell mitochondrial membrane potential and inducing accumulation of cell reactive oxygen species.
