摘 要:采用响应面法优化、超声辅助提取铁棍山药中山药素Ⅰ,结果表明最佳的提取工艺为:液料比(mL/g)46∶1、乙醇体积分数29%和超声时间63 min.Box-Behnken(BB)中心组合设计试验证明各单因素的影响顺序:乙醇体积分数>超声时间>液料比,液料比和超声时间、乙醇体积分数和超声时间的二次交互作用显著.在最佳条件下进行验证,山药素Ⅰ提取量为0.6271 mg/g,接近模型预测值0.6434 mg/g,响应面法对山药素Ⅰ提取工艺参数优化可行.Response surface methodology and ultrasonic-assisted extraction were used to optimize extraction of batatasinⅠfrom dioscorea opposita.The results showed that the best extraction process was as follows:liquid-solid ratio(mL/g)46∶1,ethanol concentration 29%and ultrasonic time 63 min.Box Behnken(BB)central composite design test showed that the order of influence of each single factor was:ethanol concentration>ultrasonic time>liquid-solid ratio.The secondary interactions between liquid-solid ratio and ultrasonic time,ethanol concentration and ultrasonic time were significant.It was verified under the best conditions that the extraction amount of batatasinⅠwas 0.6271 mg/g,close to the model prediction value of 0.6434 mg/g,and the response surface method was feasible to optimize the extraction process parameters of batatasinⅠ.
