摘 要:目的探讨聚二磷酸腺苷核糖聚合酶-1[poly(ADP-ribose)polymerase-1,PARP-1]缺陷对苯并[a]芘(benzo[a]pyrene,B[a]P)致肺癌过程的影响。方法分别以PARP-1基因敲除型、野生型小鼠作为研究对象,利用长期动式吸入B[a]P气溶胶方式染毒处理180 d,收集血液样品检测动物体内BPDE-DNA结合物含量,切取肺组织,测定其脏器系数、病理学分析检测其恶变程度。结果染毒过程中未见实验动物死亡,各组动物体重随时间逐渐增加;野生型雌性小鼠肺脏器系数明显大于对照组,差异有统计学意义(t=2.208,P<0.05),而PARP-1缺陷鼠中未见明显差异;野生型雌性小鼠肺支气管周围出现异常结节,而PARP-1缺陷鼠未见异常结节;B[a]P可诱导小鼠体内BPDE-DNA加合物形成,但PARP-1缺陷鼠体内的BPDE-DNA加合物含量相对较低(与野生鼠相比,P<0.05)。结论PARP-1缺陷可抑制B[a]P致肺癌形成。Objective To investigate the effect of poly(ADP ribose)polymerase-1(PARP-1)deficiency on lung cancer induced by B[a]P.Methods PARP-1 knockout mice and wild-type mice were treated with long-term dynamic B[a]P inhalation exposure for 180 days.Blood samples were collected to detect the content of BPDE-DNA adducts,lung tissues were taken to determine the organ coefficient and detect the degree of malignancy by pathological analysis.Results Body weight was recorded every week during the whole experiment and no animals died.The body weight of each group gradually increased over time.Lung organ coefficient of wild-type female mice after B[a]P exposure was significantly higher than that of the control(t=2.208,P<0.05),but there was no significant difference in PARP-1 deficient mice;there were abnormal lung nodules in wild-type female mice,but there were no abnormal findings in PARP-1 deficient mice;B[a]P exposure can induced the formation of induces DNA adduct in mice,but the content of BPDE-DNA adducts in PARP-1 deficient mice were relatively low(compared with wild-type mice,P<0.05).Conclusion PARP-1 deficiency can inhibit the formation of lung cancer induced by B[a]P.
