摘 要:目的探讨龙胆苦苷对CCl_(4)诱导的大鼠肝纤维化损伤模型的保护作用及机制。方法60只SD大鼠随机均分为对照组、模型组及干预组,模型组和干预组大鼠给予腹腔注射CCl_(4)诱导构建肝纤维化模型;干预组大鼠同时给予灌胃CCl_(4)干预处理。分组处理后,观察大鼠肝脏组织情况,检测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)及谷氨酰转肽酶(GGT)含量比较肝功能变化;检测学清转化生长因子-β(TGF-β)、透明质酸(HA)及胶原蛋白-Ⅲ(COL Ⅲ)含量比较肝纤维化情况;检测组织丙二醛(MDA)、超氧化物歧化酶(SOD)、羟脯氨酸(Hyp)比较氧化应激水平。结果龙胆苦苷可缓解CCl_(4)诱导引起的肝组织病变。模型组和干预组大鼠血清AST、ALT、ALP、GGT均明显高于对照组;干预组大鼠血清AST、ALT、ALP、GGT均明显低于模型组(P均<0.05)。模型组和干预组大鼠血清TGF-β1、HA、COL Ⅲ均明显高于对照组;干预组大鼠血清TGF-β1、HA、COL Ⅲ均明显低于模型组(P均<0.05)。模型组和干预组大鼠MDA和Hyp均高于对照组,SOD低于对照组;干预组大鼠MDA和Hyp均低于模型组,SOD高于模型组(P均<0.05)。结论龙胆苦苷对CCl_(4)诱导的大鼠肝纤维化损伤有明显保护作用,其机制可能与减轻氧化应激损伤相关。Objective To investigate the protective effect and mechanism of gentiopicrin on CCl_(4)induced hepatic fibrosis in rats.Methods Sixty SD rats were randomly divided into control group,model group and intervention group.Rats in model group and intervention group were given intraperitoneal injection of CCl_(4)to induce liver fibrosis model.The rats in the intervention group were treated with CCl_(4)by gavage at the same time.After treatment,the liver tissue of rats was observed,the contents of serum Alanine aminotransferase(ALT),Aspartate aminotransferase(AST),Alkaline phosphatase(ALP)and Glutamyltranspeptidase(GGT)were detected,and the changes of liver function were compared.Detection of Transforming growth factor-β(TGF-β).The contents of Hyaluronic acid(HA)and Collagen Ⅲ(Col Ⅲ)were compared with the situation of liver fibrosis.Malondialdehyde(MDA),Superoxide dismutase(SOD)and Hydroxyproline(Hyp)were detected to compare the levels of oxidative stress.Results Gentiopicrin could alleviate the liver lesions induced by CCl_(4).The serum AST,ALT,ALP and GGT in the model group and the intervention group were significantly higher than those in the control group.Serum GGT and ALT in the intervention group were significantly lower than those in the model group(P<0.05).Serum TGF-β1,Ha and col Ⅲ in model group and intervention group were significantly higher than those in the control group,and those in intervention group were significantly lower than the model group(P<0.05).MDA and Hyp in model group and intervention group were higher than those in control group,and SOD was lower than that in control group.MDA and Hyp in the intervention group were lower than those in the model group,and SOD was higher than that in the model group(P<0.05).Conclusion Gentiopicrin has a significant protective effect on CCl_(4)induced hepatic fibrosis in rats,and its mechanism may be related to reducing oxidative stress injury.
