摘 要:采用碱提醇沉法和柱层析分离法从茯苓中获得2种多糖,命名为PCAPS2和PCAPS5,其分子质量大小分别为3.97×10^(5) u和9.61×10^(5) u。采用MTT法测定PCAPS2和PCAPS5对小鼠脾细胞的增殖作用,发现2种碱提多糖均可以促进小鼠脾细胞和T淋巴细胞增殖,并且PCAPS5对脾细胞的增殖效果优于PCAPS2。利用荧光免疫标记法和抑制剂中和实验分析相关免疫机制,结果表明,PCAPS2和PCAPS5可以通过与TLR4/MR受体结合,进一步激活NF-κB以诱导巨噬细胞分泌细胞因子TNF-α。茯苓碱提多糖可以作为新型食品添加剂和免疫调节剂进一步开发利用。Two polysaccharides named PCAPS2 and PCAPS5 were obtained from Poria cocos(Schw.)Wolf by the alkali extraction and column chromatography methods.The molecular weights of PCAPS2 and PCAPS5 are 3.97×10^(5) u and 9.61×10^(5) u,respectively.The MTT assay was used to determine the effects of PCAPS2 and PCAPS5 on the proliferation of splenocytes.Both extracts enhanced the proliferation of splenocytes and T lymphocytes,and PCAPS2 was superior to PCAPS5 in proliferation efficacy.Immunofluorescence methods and inhibitor assay were used to explore the related immune mechanisms.The results showed that PCAPS2 and PCAPS5 induced the TNF-αsecretion in RAW264.7 macrophage by interacting with TLR4/MR to activate NF-κB.The polysaccharide extracted from Poria cocos(Schw.)Wolf could be further developed as new food additives and immunopotentiators.
